Non-specific Lipid Transfer Proteins were isolated and purified from maize seeds using combinations of ammonium sulphate precipitation and cat-ion exchange chromatography followed by HPLC. Two peptide segments were obtained by MALDI-TOF MS analysis, which were designated as ns-LTP M1 and ns-LTP M2. Both the ns-LTPs were treated to pearl millet seeds at different concentrations for 6 h. Among the concentrations tested, ns-LTP M1 (50 μg/ ml) showed a maximum seed germination of 94% and 1320 seedling vigor followed by ns-LTP M2. The maximum downy mildew disease protection of 62% was offered by ns-LTP M1 (50 μg/ ml) followed by ns-LTP M2 which offered 58% protection. Furthermore, peroxidase (POX) and lipoxigenase (LOX) enzymes related to plant resistance metabolism were also increased considerably after ns-LTP M1 treatment. POX activity was up to two folds when compared to susceptible control seedlings. Thus the present investigation suggests that the maize LTPs could enhance downy mildew disease resistance in pearl millet through the induction of defense mediated enzymes.

In the present study, the gene (pen2) encoding a mature Penaeidin-2 from Penaeus vannamei. shrimp and gene (rHarpinZ) encoding a HarpinZ from Pseudomonas syringae pv. tomato were respectively cloned. The pen2 was fused to rHarpinZ with a linker (GGSGSGGSSRGGSGS) and then cloned into pET-32a (+) formed the expression plasmid pET32a (+)/pen-hrp. Penhrp was expressed in the prokaryotic (E. coli Origami (DE3) pLysS) expression system. Functional analysis showed that the fusion protein Penhrp could elicit stronger hypersensitive response (HR) including higher Peroxidase (POD) and Superoxide dismutase (SOD) activities and the transcription of PR-5dB gene than the original HarpinZ. In addition, a stronger systemic acquired resistance (SAR) to TMV in Xanthi nc. tobacco was also induced. These may offer a potential effective strategy for plant disease control.

PB90 is a novel protein elicitor isolated from Phytophthora boehmeriae. Here, we report that treatment of PB90 stimulates hypericin production and hydrogen peroxide (H 2 O 2 ) generation in Hypericum perforatum L. cells and demonstrate that H 2 O 2 is essential for PB90-induced hypericin production. To further study the source of PB90-triggered H 2 O 2 , we have investigated activities of plasma membrane NADPH oxidase in Hypericum perforatum L. cells subjected to PB90 treatment. It is revealed that treatment of the cells with PB90 significantly increases NADPH oxidase activity. NADPH oxidase inhibitors suppress not only the PB90-stimulated NADPH oxidase activity but also the PB90-triggered H 2 O 2 generation and PB90-induced hypericin production, showing that NADPH oxidase is involved in PB90-triggered H 2 O 2 generation and hypericin production. Moreover, the suppression of NADPH oxidase inhibitors on PB90-induced hypericin production can be reversed by H 2 O 2 , although H 2 O 2 per se has no effects on hypericin production of the cells. Together, the data demonstrate that PB90 may induce hypericin production of H. perforatum cells through the NADPH oxidase-mediated H 2 O 2 signaling pathway.
