Purification and Characterization of Proteinase Inhibitors from Winged Bean (Psophocarpus tetragonolobus (L.) DC.) Seeds1

Abstract: Seven proteinase inhibitors were isolated from winged bean seeds by ion-exchange chromatographies. These inhibitors had molecular weights of around 20,000, included four half-cystine residues, and were Kunitz-type inhibitors. Two (WTI-2 and 3) inhibited bovine trypsin strongly and four (WCI-1, 2, 3, and 4) inhibited bovine alpha-chymotrypsin, but in different ways. One mole of WCI-2 or -3 could inhibit 2 mol of alpha-chymotrypsin. The remaining inhibitor (WTCI-1) could bind both bovine trypsin and alpha-chymot… Show more

“…The stoichiometry of ECI with chymotrypsin was estimated to be 1 :2 by the titration patterns of its inhibitory activity, suggesting that one ECI molecule binds two chymotrypsin molecules. Although similar observations were reported on chymotrypsin inhibitors from Erythrina latissima seeds [ 191, Erythrina caffra seeds [20] and winged bean [15], no further study of their interaction with chymotrypsin was made. As for 'double heads' serine proteinase inhibitors, Bowman-Birk family proteinase inhibitors which form a stable ternary complex consisting of one inhibitor and two proteinase molecules are well known [22].…”

“…Inhibiton of chymotrypsin by ECI and its derivatives was assayed, as described by Shibata et al [15], in 96-well microtiterplate. Briefly, chymotrypsin was dissolved in 1 mM HCI containing 20 mM CaCl, at the concentration of 0.1 mg/ml.…”

“…However, the technique suffered from the same serious drawback as that of Kortt that ended up with cleaved inhibitors by the cognate proteases. Again, purification method by Shibata et al [12] suffered being laborious and time consuming. However, this presently described purification method involves only a single column, is less time consuming and results in high yield of purified protein.…”

“…seeds are a rich source of Kunitz type protease inhibitors and are reported to contain at least nine trypsin or chymotrypsin inhibitors [5,12] . Its high protein and oil content has earned it a name "soybean of tropics [10] ".…”
