Purification and characterization of myrosinase from the cabbage aphid (Brevicoryne brassicae), a brassica herbivore

Abstract: Aphids are among the most serious insect pests of agricultural crops in the world. They often have specific hosts, and the cabbage aphid (Brevicoryne brassicae) is a specialist on Cruciferae. It has previously been described that certain insects contain the enzyme myrosinase (EC 3.2.3.1), which is considered an important defence enzyme of crucifers. Myrosinase was purified to homogeneity from cabbage aphid soluble extracts using anionexchange and phenyl-Sepharose chromatography. The protein has an apparent sub… Show more

“…Aliphatic glucosinolates were the preferred substrates of the flea beetle myrosinase, with aromatic and indolic glucosinolates and two different standard β-O-glucoside substrates being hydrolyzed at much lower efficiencies. The previously identified aphid myrosinase from B. brassicae (40-42) displays much broader substrate specificity (41,43).…”

“…In comparison, benzyl glucosinolate (glucotropaeolin) has been shown to have 63.3 % of the relative activity of 62 in the presence of myrosinase isolated from Brevicoryne brassicae. [80] Given that glucosinolates 38 and 39 are both significantly larger than benzyl glucosinolate and are more likely to experience space restrictions within the enzyme active site, it is not entirely surprising that these compounds act as less A C H T U N G T R E N N U N G effective myrosinase substrates than benzyl glucosinolate in comparison with 62. This is especially evident for 39, containing the highly rigid biaryl functionality.…”

“…The enzyme is localized in the non-fl ight muscles and does not show high sequence similarity to plant myrosinases; however, a homology to plant β-glucosidases could be inferred [ 190 , 192 ] and accordingly a low O -β-glucosidase activity was demonstrated using standard substrates [ 192 ]. Expression of this myrosinase increases approximately twofold throughout the developmental stages in both wingless and winged aphid forms [ 185 ].…”
