Purification and Characterization of Chitinase from Streptomyces sp. M-20

Abstract: , and p-chloromercuribenzoic acid completely inhibited the enzyme activity. The chitinase activity was high on colloidal chitin, chitotriose, and chitooligosaccharide. The purified chitinase showed antifungal activity against Botrytis cinerea, and lysozyme activity against the cell wall of Botrytis cinerea.

“…M-20 was strongly inhibited with low concentrations of metallic ions. 41) The addition of CoSO 4 at 5 mM was found to enhance the activity of the enzyme, which increased by 325% as compared to that attained without Co 2þ . This result indicates that the enzyme requires Co 2þ for optimal activity.…”

“…These enzymes have widespread applications in the fields of medicine (Kim et al 2003, Toharisman et al 2005, waste management (Odds 2003, Lopes et al 2008, biotechnology (Felse and Panda 1999a, b) and industry (Haki and Rakshit 2003). Chitinases are used for the mild degradation of chitin to prepare biologically active chitooligosaccharides and for the structural determination of the glycoprotein polysaccharide.…”
