Purification and Biochemical Characterization of a Highly Thermostable Xylanase from Actinomadura sp. Strain Cpt20 Isolated from Poultry Compost

Abstract: An extracellular thermostable xylanase from a newly isolated thermophilic Actinomadura sp. strain Cpt20 was purified and characterized. Based on matrix-assisted laser desorption-ionization time-of-flight mass spectrometry analysis, the purified enzyme is a monomer with a molecular mass of 20,110.13 Da. The 19 residue N-terminal sequence of the enzyme showed 84% homology with those of actinomycete endoxylanases. The optimum pH and temperature values for xylanase activity were pH 10 and 80 °C, respectively. This… Show more

“…Likewise, the activity of Actinomadura sp. strain Cpt20 xylanase was also found to be enhanced by Ca 2+ and Co 2+ , but was inhibited by Fe 2+ , Zn 2+ , Cu 2+ and Mg 2+ , yet almost unaffected by K + , Na + and Mn 2+ [24]. In contrast to the results obtained, ethanol, isopropanol and butanol had no effect on the Streptomyces rameus L2001 xylanase activity [26].…”

“…However, a few organisms show constitutive production of the enzyme and also catabolic repression by carbon sources such as glucose or xylose (Walia et al 2013a, c). Xylan has been shown to be the best inducer of xylanase production in many cases (Taibi et al 2012; Guleria et al 2013; Walia et al 2014). Xylan, being a high molecular mass polymer, cannot enter the cell.…”

“…Such specifications relate to costeffectiveness, eco-friendliness, and ease of use (Taibi et al 2011). In this paper we reported kinetics of partially purified xylanases from T. harzianum and C. thermophilum ATCC 28076 under optimized culture conditions.…”
