Purification and Application of a Lipase from Penicillium expansum PED-03

Abstract: An extracellular lipase was purified from the fermentation broth of Penicillium expansum PED-03 by DEAE-Sepharose chromatography, followed by sephacryl S-200 chromatography. The enzyme was purified 81.8-fold with 19.8% recovery and a specific activity of 85.94 U/mg. The molecular weight of the homogeneous enzyme was about 28 kDa, determined by sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis. The enzymatic resolution of racemic ibuprofen was carried out by the lipase from P. expansum PED-03, and… Show more

“…The SDS-PAGE results strongly suggest that PCrL is a monomeric protein comparable to those previously reported for other lipases from Penicillium strains. The molecular mass of PCrL (28 kDa) is similar to that of the Penicillium expansum strain PED-03 [ 39 ]. Most of the known lipases from the genus Penicillium have been reported to have a molecular mass in the range of 25–43 kDa [ 22 , 40 , 41 ].…”

“…The range of substrates converted by lipases can be learned from the following examples. In the work by Lianghua and coworkers, ibuprofen in R and S racemate reacts with 1-propanol in the presence of lipase from Penicillium expansum [23] while ( S )-ibuprofen n-propylester and ( R )-ibuprofen are the products. The reaction can be applied for the preparation or manufacturing of an optically pure isomer of ibuprofen.…”

“…Thus, screening and using expression signals of other highly expressed genes in this fungus will definitely be a promising approach to further promote the performance of this engineered strain. P. expansum is an economical important filamentous fungus in consequence of its content of alkaline lipase, which is now widely used in many fields including biodiesel [31], detergent [32], leather [33], and fine chemicals [34]. Based on the newly characterized Ppel, the overexpression vector was constructed and after transformation and screening, recombinant strains that showed a significant increase (P < 0.05) in lipase yield were successfully obtained, which makes our research practically significant.…”
