Pumilicin 4, A Novel Bacteriocin with Anti-MRSA and Anti-VRE Activity Produced by Newly Isolated Bacteria Bacillus pumilus Strain WAPB4

Abstract: A total of 34 bacterial strains with anti-methicillin-resistant Staphylococcus aureus (MRSA) activity were isolated from 69 soil and water samples collected from four areas of Thailand. One strain, WAPB4 identified as Bacillus pumilus, showed remarkable antibacterial activity against MRSA, vancomycin-resistant Enterococcus faecalis (VRE), and several Gram-positive test bacteria. Bacteriocin produced by WAPB4 was designated as pumilicin 4. It was heat stable up to 121 degrees C, 15 min and active within the pH … Show more

“…The antibacterial activity was reduced to 25% and 50% at pH 2 and 3, respectively, and was severely altered at pH higher than 10 ( Table 2). The heat-stable property was also observed in other antimicrobial metabolites produced by Bacillus species [14,15,38,39]. The stability of this antibacterial activity over a wide range of pH and after heat treatment might be useful in several industrial applications.…”

“…Only few antimicrobial compounds produced by the genus Bacillus exhibit inhibitory activity against methicillin-resistant Staphylococcus species [15]. The antimicrobial activity against methicillin-resistant S. aureus species has only been described for Bacillus subtilis producing macrolactin antibiotic [3] and Bacillus pumilis strain producing pumilicin 4 [15].…”

“…This bacterium is highly resistant to extreme environmental conditions such as low or no nutrient availability, desiccation, irradiation, H 2 O 2 and chemical disinfections (19). The ecological role of B. pumilus is emphasized by the fact that they do produce compounds antagonist to fungal and bacterial pathogens (4,6). Thus, B. pumilus is of considerable research interest to understand its physiological diversity, genetic relatedness with other Bacillus spp.…”

“…Some B. pumilus strains show fungicidal activity and have been used as biological control agents against phytopathogenic fungi (9,42). Other B. pumilus strains have been reported to be plant growth-promoting rhizobacteria (25,36,57,77), while some others showed potent antibacterial activity (6). Still others have been used as probiotics (15,20,28).…”
