The influence of Ascaris extract, applied during 1 h in low concentration as aerosol and intravenously, on the bronchial system and the potentiation of increase of airway resistance to acetylcholine (ACH) and histamine challenge caused by this kind of allergen exposure were studied in 13 dogs. All the animals presented no reaction by the allergen exposure itself but developed an increased response to ACH and histamine after this kind of allergen exposure. A control group of 4 dogs was submitted to saline solution aerosol and intravenous application. No change in response to the substances mentioned was observed. The airway response to cold air after concentrated and diluted Ascaris suum extract (AE) aerosol application was tested on 7 dogs. The response to cold air was only influenced after concentrated AE-aerosol exposure.

A B S T R A C T Measurements of respiratory mechanics, arterial blood gases, and pulmonary vascular resistance were made before and 15 min after inhalation challenge with Ascaris suum extract in dogs with natural sensitivity to this antigen. 25 of 47 dogs were treated before inhalation challenge with a prostaglandin inhibitor (90 mg/kg of aspirin or 2 mg/kg of indomethacin by intravenous infusion). In response to the challenge, bronchospasm developed in approximately half (responders) of each group reflected by decreases in mean specific respiratory system conductance and arterial oxygen tenision. While the dogs were breathing room air, pulmonary vaseular resistance remained unchanged after antigen challenge in the responders not given aspirin or indomethacin, but increased significantly and was associated with a lesser degree of arterial hypoxemia in the responders pretreated with either of the prostaglandin inhibitors. Prevention of arterial hypoxemia by oxygen breathing blocked an increase in pulmoinary vascular resistance in four pretreated responiders. No changes in respiratory mechanies, pulmonary hemodyiianaies, or arterial blood gases were noted in the 21 dogs who did not develop bronchospasmi regardless of whether or not they wvere pretreated. 12 additional dogs in whom arterial hypoxemia was produiced by 10% oxygen breathing, showed an increase in pulmonary vaseular resistance that was not potentiated by pretreatment with aspirin in 6. We conclude that in acuite experimental canine asthma, vasodilator prostaglandins appear to blunt the hypoxic pulmonary vasoconstrictor response, thereby further compromising gas exchange btut preventing the development of pulmonary hypertension.

Because of the public concern surrounding the issue of the safety of genetically modified organisms, it is critical to have appropriate methodologies to aid investigators in identifying potential hazards associated with consumption of foods produced with these materials. A recent panel of experts convened by the Food and Agriculture Organization and World Health Organization suggested there is scientific evidence that using data from animal studies will contribute important information regarding the allergenicity of foods derived from biotechnology. This view has given further impetus to the development of suitable animal models for allergenicity assessment. This article is a review of what has been achieved and what still has to be accomplished regarding several different animal models. Progress made in the design and evaluation of models in the rat, the mouse, the dog and in swine is reviewed and discussed.
