It is accepted that an effective prophylactic HIV-1 vaccine is likely to have the greatest impact on viral transmission rates. As previous reports have implicated DNA-priming, protein boost regimens to be efficient activators of humoral responses, we sought to optimize this regimen to further augment vaccine immunogenicity. Here we evaluated single versus concurrent intradermal Using an alternative and larger animal model, the rabbit, we found the concurrent DNA-priming strategy followed by s.c. protein boosting to again be capable of eliciting high-avidity humoral responses and to also be able to neutralize HIV-1 pseudoviruses from diverse clades (clades A, B, and C). Taken together, we show that concurrent multiple-route DNA vaccinations induce strong cellular immunity, in addition to potent and high-avidity humoral immune responses. IMPORTANCEThe route of vaccination has profound effects on prevailing immune responses. Due to the insufficient immunogenicity and protection of current DNA delivery strategies, we evaluated concurrent DNA delivery via simultaneous administration of plasmid DNA by the i.m. and i.d. routes. The rationale behind this study was to provide clear evidence of the utility of concurrent vaccinations for an upcoming human clinical trial. Furthermore, this work will guide future preclinical studies by evaluating the use of model antigens and plasmids for prime-boost strategies. This paper will be of interest not only to virologists and vaccinologists working in the HIV field but also to researchers working in other viral vaccine settings and, critically, to the wider field of vaccine delivery.

Purpose: To evaluate the comparative effect of topical versus intramuscular administration of nanoparticle-carried DNA vaccine in preventing corneal herpes simplex virus type 1 (HSV-1) infection. Methods: Nanoparticle [polyethylenimine (PEI)-Fe₃O₄]-carried DNA vaccine (PEI-Fe₃O₄-pRSC-gD-IL-21) or DNA vaccine (pRSC-gD-IL-21) alone were topically versus intramuscularly inoculated into one eye each of mice on days 0, 14 and 28. Three weeks after the final immunization, the specific immune responses and clinical degrees of primary herpes simplex keratitis were evaluated. Results: Topical inoculation of nanoparticle-carried DNA vaccine induced mice to generate similar levels of specific HSV-1-neutralizing antibody, IFN-γ and IL-4 in serum and specific killing (cytotoxicity) and proliferative activities of the splenic lymphocytes, but a significantly higher level of secretory IgA in tears compared to those of intramuscular inoculation. More importantly, the mice inoculated topically showed a significantly decreased herpes simplex keratitis severity than the mice inoculated intramuscularly after HSV-1 challenge on the corneas of the mice. Conclusions: Topical inoculation of nanoparticle-carried DNA vaccine elicits a stronger specific local immune response and more effectively inhibits herpes simplex keratitis as compared to intramuscular inoculation in an HSV-1 ocular challenge mouse model. Thus, topical administration may be a promising inoculating route for the nanoparticle-carried DNA vaccine to prevent corneal infections.

Shaji and Shaikh: Current Development of Pulmonary Drug DeliveryThe pulmonary drug delivery system offers several merits over other drug delivery systems and therefore, this delivery route has been in prime focus for various applications like local and systemic therapeutics delivery. The overall development of drug delivery system depends on its efficacy, quality and safety and to achieve such attributes there is a need of reliable evaluation methods to test them. This review provides an in-depth analysis of the development in the evaluation of pulmonary drug delivery systems. In vitro methods of testing pulmonary products such as particle morphological studies, powder flow characteristics, moisture content test, aerosol turboelectric characterization, particles interparticulate forces measurement and solid state characterizations were discussed. Particle size and zeta potential measurement and evaluation of aerosol performance such as dose uniformity and aerodynamic particle size distribution were reviewed in detail. The development of dissolution methods for pulmonary products is also elaborated. Various cell culture methods for testing pulmonary products were overviewed. The in vivo testing methods including drug administration systems, drug deposition studies and pharmacokinetic studies and ex vivo testing models were also highlighted. Together an overview of current advancement in evaluation and characterization of pulmonary drug delivery system can be analyzed and studied through this review.
