Puerarin inhibits TRPM3/miR‐204 to promote MC3T3‐E1 cells proliferation, differentiation and mineralization

Abstract: Puerarin is an isoflavonoid phytoestrogen extracted from the root of Radix Pueraria, has attracted increasing attention because of its beneficial effects on anti-osteoporosis, but the molecular mechanisms underlying its actions on osteoblasts are not fully understood. The current study aimed to investigate the effect of puerarin on MC3T3-E1 osteoblastic cells proliferation, differentiation and mineralization, in vitro and its underlying mechanisms. The results indicated that puerarin significantly promoted the… Show more

“…Puerarin([8‐(β‐ d ‐glucopyranosyl‐7‐hydroxy‐3‐(4‐hydroxyphenyl)‐4H‐1‐benzopyran‐4‐one]) is a major bioactive isoflavone compound isolated from the roots of the Pueraria lobata and has been clinically used for its protection against inflammation, oxidative stress, and mitochondrial dysfunction (Wang, Shi, et al, ; Zeng et al, ). It is reported that puerarin might be beneficial to improve lipid metabolism (Prasain, Peng, Rajbhandari, & Wyss, ), restore mitochondrial function (Song, Liu, Wang, & Wang, ), and meliorate glucose homeostasis (Yang et al, ).…”

“…Puerarin, an isoflavone extracted from the Chinese medicine pueraria, exhibits protective functions on the cardiovascular system, nervous system, osteoporosis, liver injury, and inflammation in vivo and in vitro (4). Studies have shown that puerarin can promote the proliferation, differentiation and mineralization of osteoblasts in vitro (5,6). In a recent report, puerarin was found to improve bone loss in estrogen-deficient rats, display a superior anti-osteoporosis effect, and prevent osteoporosis in postmenopausal women (7).…”

“…To date, LC3B has been extensively studied and identified as an autophagic membrane-associated factor (13). Previous studies have demonstrated that a low concentration of puerarin can promote the viability and differentiation of osteoblastic MC3T3-E1 cells, which may be achieved by downregulation of the expression of miR-204 and its host gene TRPM3 (6). However, it is not clear whether there are other targets in this pathway.…”
