Puerarin inhibits the development of osteoarthritis through antiinflammatory and antimatrix‐degrading pathways in osteoarthritis‐induced rat model

Abstract: Puerarin is an isoflavone isolated from the medicinal plant Pueraria lobata. The purpose of this study was to study the antiinflammatory and antimatrix-degrading effects of puerarin in a rat osteoarthritis (OA) model and its protective effects on joints. The rat OA model was established by anterior cruciate ligament transection (ACLT) surgery. Rats (n = 40) were divided into nontreated OA, OA + celecoxib (2.86 mg/kg), OA + puerarin (50 and 100 mg/kg), and control groups. Two weeks after surgical induction, puer… Show more

“…The rats were randomly divided into five groups: control (sham operation and treated with a salt solution), OA (ACLT surgery to induce OA), celecoxib (2.86 mg/kg/day celecoxib [Pfizer Pharmaceutical, New York, United States] ( Panahifar et al, 2014 ) was administered as a positive control), 5 mg/kg + Bilobalide (the OA rats treated with 5 mg/kg/day of bilobalide), and 10 mg/kg + Bilobalide (the OA rats treated with 10 mg/kg/day of bilobalide), with eight rats in each group. The ACLT-induced PTOA models for rats and animal postoperative care were established according to previous studies ( Ma et al, 2020 ). Rats in the 5 mg/kg + Bilobalide and 10 mg/kg + Bilobalide groups received oral bilobalide daily for 6 weeks.…”

“…Pueraria lobata is widely used in traditional Chinese medicine to treat gastrointestinal diseases, because of its excellent curative effects. Puerarin, an isoflavone component purified from P. lobata, possesses an extensive spectrum of biological activities, including anti-inflammatory, anti-tumor, antiapoptosis, and immunomodulatory (Li et al, 2018;Ma et al, 2020). Puerarin injected intraperitoneally has a high absorption level, may be absorbed by the intestine in a short time, and is not metabolized into other compound forms (Yamamoto et al, 2019).…”

“…Prior to surgery, the rats were acclimated for 1 week. In the present study, an anterior cruciate ligament transection (ACLT) model was established in which rats developed OA posttrauma, using a protocol described in a previous study [23] (Figures 1(a)-1(e)). The rats were randomly allocated into a control group (sham surgery), OA group (ACLT surgery that induced OA), 20 mg/kg emodin group, 50 mg/kg emodin group, or an 80 mg/kg emodin group, each group having 8 rats.…”
