Long non-coding RNA (lncRNA) MALAT1 has been confirmed to function as an oncogene in various solid tumors. MALAT1 level has been shown to be upregulated in relapsed acute lymphoblastic leukemia (ALL) patients, but the mechanism is unclear. This study aims to investigate the functional roles and underlying mechanisms of MALAT1 in ALL. MALAT1 and miR-205 expression were assessed by real-time quantitative polymerase chain reaction (RT-qPCR). MTT assay and flow cytometry were performed to evaluate cell proliferation and apoptosis, respectively. Protein level of protein tyrosine kinase-7 (PTK7) was detected by Western blot assay. Dual luciferase reporter assay was conducted to confirm the binding of MALAT1 and miR-205, as well as miR-205 and PTK7. The levels of MALAT1 and PTK7 were upregulated in ALL samples. In contrast, miR-205 level was downregulated in ALL in ALL samples. Moreover, MALAT1 silencing or miR-205 overexpression restrained proliferation and promoted apoptosis of ALL cells. Mechanistically, MALAT1 sponged miR-205 to regulate PTK7 expression. In summary, MALAT1 affected ALL cell proliferation and apoptosis via regulating miR-205-PTK7 axis. Our results suggest that MALAT1-miR-205-PTK7 axis participates in the proliferation and apoptosis of ALL, which may provide a potential treatment target for ALL.

Oral squamous cell carcinoma of the tongue is the most common type of cancer, common site being buccal mucosa and gingiva. Despite advancement in treatment protocol, recurrence of the disease limits outcome of oral cancer; contributing factors for poor prognosis are activation, overexpression, dysregulation or mutation of many target proteins including protein tyrosine kinases (PTKs). Therefore, it is time to develop preventive measures against oral carcinogenesis, particularly by regulating tyrosine kinases. Natural compounds find a place in cancer prevention; hence, emphasis has been given to investigate the chemopreventive efficacy of black tea extract in 4-nitroquinoline-1-oxide (4NQO)-induced tongue tumorigenesis in C57BL/6 mice. The histological diagnosis of squamous neoplasia was observed at different time intervals in mice treated with 4NQO and tea group, alone or in conjunction. Results indicate the preventive role of black tea in 4NQO-induced lesions. Expressions of different tyrosine kinases, their phosphorylated forms (EGFR, JAK-1, JAK-2, p38 MAPK) and their downstream signalling proteins (PI3K, Akt, Raf1 and Stat3) were determined by western blot analysis at different stages of oral carcinogenesis. 4NQO-induced endogenous ROS generation level and corresponding DNA damage by comet assay were followed in presence and absence of tea. An upward trend of ROS level as well as DNA damage was observed with progression of oral carcinogenesis. Administration of black tea extract inhibited tongue tumorigenesis by down-regulation of protein tyrosine kinases, reducing ROS level and counteracting DNA damage. Black tea therefore exhibited efficient chemo-preventive activity in oral carcinogenesis.

Background: Protein tyrosine kinase-7, a regulatory protein in the Wnt signaling pathway, was highly overexpressed in various cancer types and assumed to be related to prognosis. Aims: The purpose of this study is to assess whether protein tyrosine kinase-7 expression status in curatively resected gastric carcinoma would independently identify patients with a high risk of recurrence and death. Study Design: Retrospective cohort study. Methods: We included patients who were at least 18 years of age and diagnosed with gastric cancer. The exclusion criterion was a metastatic disease at the time of diagnosis or operation. Data on clinicopathological prognostic determinants and clinical courses, including the date of disease relapse and survival status, were collected with the use of medical records. Surgically removed tumor tissue specimens were examined by two independent pathologists at the pathology department of our institution. Protein tyrosine kinase-7 expression status was assessed with immunohistochemical processing and stratified on a scale ranging from 0 to +3 according to the extent of stained tumor cells. It was then further categorized into two groups, one being + (positive), including +1, +2, and +3 scores, another was-(negative), including-and +/− scores. Results: A total of 114 patients were analyzed. Protein tyrosine kinase-7 expression was present in 66.7% of the surgical tumor specimens. There was no statistically significant difference in almost all relevant parameters between the protein tyrosine kinase-7 positive and negative groups. The estimated median survival in the protein tyrosine kinase-7 positive group was significantly better than the protein tyrosine kinase-7 negative group (60 vs 22 months, p<0.001). Disease-free survival was found to be 55 months in the protein tyrosine kinase-7 positive group, whereas it was 21 months in the negative group (p=0.015). In the multivariate analysis, along with negative protein tyrosine kinase-7 expression, poor performance status, and advanced stage were significantly associated with the risk of death (p<0.001 for each). Conclusion: Compared to patients with negative PTK-7 expression, patients with positive PTK-7 expression have better disease-free survival and overall survival rates. Efforts should be made to enhance this finding and translate it into clinical practice.
