PRRX1 Is a Novel Prognostic Biomarker and Facilitates Tumor Progression Through Epithelial–Mesenchymal Transition in Uveal Melanoma

Abstract: Uveal melanoma (UM) is the most common primary intraocular malignancy in adults. UM develops and is sustained by inflammation and immunosuppression from the tumor microenvironment (TME). This study sought to identify a reliable TME-related biomarker that could provide survival prediction and new insight into therapy for UM patients. Based on clinical characteristics and the RNA-seq transcriptome data of 80 samples from The Cancer Genome Atlas (TCGA) database, PRRX1 as a TME- and prognosis-related gene was iden… Show more

“…At present, studies of UVM have elucidated many mechanisms and found new predictors and potential targets (62)(63)(64), but no specific mechanism of the B7 family has been studied. To explore the mechanism of B7 family, we performed GO and KEGG analysis.…”

“…Peculiarly, micrometastatic lesions were devoid of CSCs, but on the other hand, de novo CSCs were indeed present in overt metastases [ 55 ]. After extravasation, metastatic colonization is further stimulated by MET, which triggers the re-expression of E-cadherin and the downregulation of Prrx1, which subsequently decreases Snail 1 expression and leads to the re-acquisition of an epithelial phenotype and epithelial morphology [ 56 , 57 ].…”

“…Paired related homeobox 1 (PRRX1), is a key member of the homomeric protein pairing family located at the nucleus. PRRX1 mediates cancer cell invasion and metastasis by starting EMT ( Meng et al, 2022 ). In addition, PRRX1 impacts the division and metastasis of various tumor cells via Wnt/β-catenin and Notch pathways, and maintains the characteristics of tumor stem cells to promote EMT ( Du et al, 2021 ).…”
