Provision of antigen and CD137 signaling breaks immunological ignorance, promoting regression of poorly immunogenic tumors

Abstract: Treatment of advanced, poorly immunogenic tumors in animal models, considered the closest simulation available thus far for conditions observed in cancer patients, remains a major challenge for cancer immunotherapy. We reported previously that established tumors in mice receiving an agonistic mAb to the T cell costimulatory molecule 4-1BB (CD137) regress due to enhanced tumor antigen–specific cytotoxic T lymphocyte responses. In this study, we demonstrate that several poorly immunogenic tumors, including C3 tu… Show more

“…Potential mechanisms of 4-1BB agonistic antibody-mediated tumor regressions include breaking of immunologic ignorance and prevention of T-cell deletion and anergy (5,35,36). Given that 4-1BB is specifically expressed on antigen-experienced T cells (7,37), the observed increase in 4-1BB þ T cells may implicate an enhanced immune response to tumor challenge in anti-4-1BB-treated animals.…”

“…15 4-1BB mediates signaling either by ligation with natural ligand expressed on activated APC or experimentally by binding an anti-4-1BB agonistic antibody. We previously developed a recombinant adenovirus expressing membrane-bound mouse 4-1BBL (ADV/4-1BBLm) consisting of the extracellular domain and the transmembrane domain of 4-1BBL.…”
