Proviral load and immune function in blood and lymph node during HIV-1 and HIV-2 infection

Abstract: SUMMARYProviral load as well as lymphocyte phenotype and function were compared in peripheral blood and lymph node compartments of 17 HIV-1, 12 HIV-2 and three dually infected patients with lymphadenopathy. The mean percentage (95% confidence interval (CI)) of CD4 þ cells was higher in lymph node mononuclear cells (LNMC) than in peripheral blood mononuclear cells (PBMC) in both infections, being 26·7% (21·1%, 32·3%) and 15·3% (10·4%, 20·2%), respectively, for HIV-1-infected patients (P ¼ 0·0001) and 32·3% (22·… Show more

“…There are no data available on lymphoid tissues during HIV-2 disease. Of note, the establishment of HIV-2 infection is associated with levels of proviral DNA similar to those found in HIV-1, suggesting an equivalent viral dissemination despite the reduced HIV-2 viremia [26][29],[72]. Therefore, it is plausible that HIV-2 RNA and/or HIV-2 proteins may induce pDC maturation and migration.…”

“…Levels of proviral DNA are similar in both infections (although there may be differences in proviral DNA levels between HIV‐1 and HIV‐2 if more sophisticated analytical methods are used ), and viral RNA is usually undetectable in aviraemic HIV‐2‐infected subjects, despite demonstrable proviral DNA . Within cohorts, variations in proviral DNA correlate with plasma viral load (and rates of CD4 decline) for both infections but the relationship between proviral DNA and circulating viral RNA is quite different for the two viruses . Interestingly, proviral load in peripheral blood cells correlates closely with that in lymph node lymphocytes suggesting that infected cells circulate freely between nodes and blood .…”

“…If the RNA viral loads in HIV-1 and HIV-2 infections differ significantly, the proviral loads in the peripheral blood are similar (5,20), and they are approximately five times higher in lymph nodes for both infections (14). Thus, a number of circulating CD4 T cells in infected individuals carry the HIV-2 provirus and may express the nef gene independently of the functionality of the other viral genes.…”
