SUMMARYThe objectives of the present study were to examine proventriculi of broiler chicks for lesions, and to classify and record the incidence of these lesions.Deep non-purulent necrotizing proventriculitis (accompanied by adenoepithelial hypertrophy and hyperplasia) was the most common (109/220 = 49.5%) light microscopic diagnosis in the proventriculi examined. Degenerating and necrotic alveolar secretory cells had amorphous, granular or vacuolated cytoplasm. Nuclei usually were either pyknotic, karyorrhectic or karyolytic; however, fewer attached or sloughed cells had swollen nuclei with marginated chromatin and clear centres. Basophilic inclusion bodies were never seen. In five cases examined ultrastructurally, hexagonal virus particles were found in intact nuclei (average size = 68.9 nm, n = 89), associated with unbound condensed chromatin, and within vacuolated spaces in the cytoplasm (average size = 62.3 nm, n = 109). DNA in situ hybridization failed to detect adenovirus or polyomavirus nucleic acids.The presence of intralesional virus suggests that a causal relationship might exist between the virus and the proventricular lesions.

“…In Australia, REV was first isolated from a duck with lymphoreticular cell tumours by Grimes and Purchase (1973). Attention was subsequently drawn to the pathogenic potential of REV in chickens when widespread losses occurred among Australian poultry flocks (Jackson et al, 1977) following the use of a commercial Marek's disease vaccine which was subsequently proven to be contaminated with REV . A recent study by Grimes et al (1979) showed that infection of young chickens with an Australian isolate of REV resulted in feathering defects and poor growth rate, haematological changes, hypoplasia of organs of the immune system and inflammation in visceral organs and the nervous system.…”

SUMMARYFifteen SPF chickens were inoculated with an Australian strain of reticuloendotheliosis virus (REV) at 1 day of age and five uninoculated chickens were readily infected by horizontal spread from this group. Antibody detectable by the immunofluorescent antibody (IFA) test developed 3 to 6 weeks after infection, and usually persisted for 20-35 weeks, with maximum titres (40-1280) at 8 to 13 weeks. Agar gel precipitin (AGP) reactions developed more slowly and were variable in duration, the highest proportion of positive reactions being detectable 8 to 13 weeks after infection and persisting for 8 to 30 weeks.Infectious REV was readily detected in the plasma and serum of inoculated chickens 6 weeks after infection and a non-infectious REV antigenaemia usually persisted for at least a further 7 weeks, in the presence or absence of antibody. Development of a detectable REV viraemia was strongly associated with poor body development and premature mortality among the inoculated chickens. In two inoculated chickens which failed to develop detectable serological reactions, a REV viraemia occurred which persisted throughout life. At autopsy, REV was re-isolated from the kidneys of most of the inoculated chickens and from the reproductive and intestinal systems of two birds 22 and 56 weeks after infection.

Detection of reticuloendotheliosis virus infection using the polymerase chain reaction

SUMMARYThe polymerase chain reaction (PCR) was shown to be a sensitive and useful method for detection of infection by members of the group of avian reticuloendotheliosis virus (REV). Genomic DNA extracted from chick embryo fibroblasts (CEFs), blood and tumours of chickens experimentally infected with the spleen necrosis virus (SNV) strain of REV was used as the target for chain elongation. Deoxyoligonucleotide primers that encompass a portion of the unique 3', repeat and unique 5' region of the long terminal repeat (LTR) served to prime chain elongation. Products characteristic of the SNV LTR were also produced from DNA extracted from CEF infected with other strains of REV, namely chick syncytial virus, duck infectious anaemia virus, and the T-strain of REV. SNV-LTR sequences were amplified from DNA of SNV-experimentally infected chicks between 5 days and 19 weeks after infection. SNV-LTR sequences were also amplified from DNA from tumours and brains of SNV infected chickens, but not from DNA from avian leukosis virus-or Marek's disease virus-induced tumours. Results indicate that PCR is a sensitive and specific method for detection of REV infection and tumours.
