“…In most cases of CL, microscopy with Giemsa staining at pH 6.8 of smears from aspirates, scrapings or biopsy specimens can detect Leishmania amastigotes within macrophages in most cases. 52 However, as lesions of CL age, there is a gradual decrease in the number of amastigotes found in parasitized macrophages and a progressive increase in tuberculoid granulomas, 53 explaining the absence of detectable amastigotes in this patient's biopsy specimens. PCR has emerged as a new tool in the diagnosis of Leishmania and has been shown to be more sensitive than microscopic examination and culture methods.…”
