Protocol for dissection and culture of murine dorsal root ganglia neurons to study neuropeptide release

Abstract: Summary In this protocol, we provide step-by-step instructions for dissection and culture of primary murine dorsal root ganglia (DRG), which provide an opportunity to study the functional properties of peripheral sensory neurons in vitro . Further, we describe the analysis of neuropeptide release by ELISA as a possible downstream application. In addition, isolated DRGs can be used directly for immunofluorescence, flow cytometry, RNA sequencing or proteomic approaches, electrop… Show more

“…DRG were dissected from naïve Swiss mice, digested, and seeded on Nunc™ Glass Bottom Dishes at a density of 1 × 10 6 per mL (1 × 10 5 /plate). Plates were incubated overnight in neurobasal medium, as previously described [ 38 , 52 ]. Neurons were treated either with vehicle (2% DMSO in culture media) or KA (50 and 150 μM) for 1 h at 37 °C and 5% CO 2 atmosphere.…”

“…Animals were killed by CO 2 inhalation, followed by cervical dislocation and postmortem decapitation. Dissociated DRG cultures were prepared as previously described (Perner & Sokol, 2021 ). Briefly, DRGs were dissected from 1‐ to 2‐month‐old mice in Hank's buffered salt solution (HBSS) and digested with Collagenase A and Dispase II (Sigma‐Aldrich) for 25 min at 37°, centrifuged at 200× g for 5 min and washed with Dulbecco's modified Eagle's medium (DMEM) supplemented with 10% fetal bovine serum (FBS), glutamate, sodium pyruvate, 1% penicillin and 1% streptomycin.…”

“…Primary DRG neurons were prepared from 8 to 10-week old mice as described previously [ 28 ]. Briefly, lumber DRG (L1 to L6) were isolated from mice and incubated with Dispase-II Solution (Sigma-Aldrich, St. Louis, MO, USA) and collagenase type II (Worthington, Columbus, OH, USA) for 70 min and with 0.25% trypsin for 5 min.…”
