Protocatechuic acid activates key components of insulin signaling pathway mimicking insulin activity

Abstract: This study showed that PCA stimulates the insulin signaling pathway in human adipocytes increasing GLUT4 translocation and glucose uptake. Decreasing insulin resistance is a most desirable aim to be reached for an effective therapeutic/preventive action against metabolic syndrome and type 2 diabetes. Identifying specific food/food components able to improve glucose metabolism can offer an attractive, novel, and economical strategy.

“…Actually, serine-302 phosphorylation of IRS-1 is reported to be an obvious indicator of IR and its enhancement often occurred in hepatic IR, as we have shown in the present study ( Fig. 6A and G) [39]. However, the long-term administration of LJP61A ameliorated IR by reducing serine-302 phosphorylation of IRS-1 in LDLr −/− mice fed HFD (Fig.…”

“…Significant circulating metabolites include PCA and its metabolites vanillic and hippuric acid, as well as phloroglucinaldehyde and its metabolite ferulic acid (69,100) . In fact, some of these metabolites have been shown to be important for glucose uptake, insulin signalling and adipokine expression in in vitro studies in human and rat adipocytes (101)(102)(103) . Diabetic KK-A y mice subjected to 5-week daily administration of concentrated bilberry extract (27 g/kg diet) showed a marked reduction in fasting plasma glucose and improved insulin sensitivity (104) .…”

“…activates key components of insulin signalling pathways (Scazzocchio et al, 2015) and is protective of ischaemia/ reperfusion induced injuries of the rat heart (Tang et al, 2014) and mouse intestine (Ma et al, 2014). There is little information on the health effects of ρ-hydroxybenzoic acid other than the role of their alkyl esters, or parabens, which have been depicted as having androgen antagonist-type activity (Darbre & Harvey, 2008).…”
