Prothymosin α and a prothymosin α-derived peptide enhance TH1-type immune responses against defined HER-2/neu epitopes

Abstract: BackgroundActive cancer immunotherapies are beginning to yield clinical benefit, especially those using peptide-pulsed dendritic cells (DCs). Different adjuvants, including Toll-like receptor (TLR) agonists, commonly co-administered to cancer patients as part of a DC-based vaccine, are being widely tested in the clinical setting. However, endogenous DCs in tumor-bearing individuals are often dysfunctional, suggesting that ex vivo educated DCs might be superior inducers of anti-tumor immune responses. We have p… Show more

“…PTMA is a highly acidic protein, that is, expressed both in the nucleus and cytosol of astrocytes (Halder & Ueda, 2012). Similar to C3, PTMA is also involved in propagation of inflammatory signals via its C-terminal domain that stimulates pro-inflammatory responses and cytokine expression through activation of toll like receptor 4 (Gusella, Teixeira, Aberg, Uversky, & Mosoian, 2016;Ioannou et al, 2013;Mosoian et al, 2010). LOX is an amine oxidase that converts primary amines into aldehydes generating hydrogen peroxide and ammonium as byproducts (Williamson & Kagan, 1987).…”

“…tumor-specific T-lymphocyte response, interleukin-2 receptor expression or T[H]1-type immune responses). [10][11][12][13][14][15] Recently, two new PTMA variants (iso b and pseudogene 7) lack of nuclear localization signal (NLS) were isolated and identified from CD8 + T cells and cervicovaginal lavage with potent anti-HIV-1 activity, which were suggested to be innate immune mediators involved in immune surveillance. [16][17][18][19][20] PTMA can induce the import of Kelch-like ECHassociated protein 1 (Keap1) into the nucleus to inhibit nuclear factor erythroid 2-related factor 2 (Nrf2)-mediated antioxidant response through Nrf2 degradation.…”

“…Prothymosin alpha (ProTα), a nuclear protein, is implicated in multiple functions including cell survival, proliferation and immunogenicity (Jiang et al, 2003;Mosoian, 2011;Ueda et al, 2012;Cannavo et al, 2013;Emmanouilidou et al, 2013;Ioannou et al, 2013;Chang et al, 2014;Lin et al, 2015). We previously identified ProTα as an anti-necrotic factor in the conditioned medium of a serum-free primary culture of cortical neurons (Ueda et al, 2007;Ueda, 2009).…”
