Proteomics analysis reveals a role forE. colipolyphosphate kinase in membrane structure and polymyxin resistance during starvation

Kanchi Baijal,
Iryna Abramchuk,
Carmen M. Herrera
et al.

Abstract: Polyphosphates (polyP) are chains of inorganic phosphates that can reach over 1000 residues in length. In Escherichia coli, polyP is produced by the polyP kinase (PPK) and is thought to play a protective role during the response to cellular stress. However, the molecular pathways impacted by PPK activity and polyP accumulation remain poorly characterized. In this work we used label-free mass spectrometry to study the response of bacteria that cannot produce polyP (∆ppk) during starvation to identify novel path… Show more

“…Moreover, exposure to reactive oxygen and chlorine species (34, 74), osmotic imbalances, low pH (75), and elevated temperatures (76) induce polyP production. It is therefore not surprising that ppk1 -deficient cells are characterized by pleiotropic phenotypes, such as increased susceptibility towards oxidative stress (33, 34, 77), antibiotics (78), elevated temperatures (76, 77), heavy metals (25), lipid A modification (78), and starvation (78). In addition, various studies point towards high sensitivity of bacteria towards changes in polyP metabolism given that both a reduction in the cellular polyP level (i.e.…”
