Proteomic Study Related to Vascular Connections in Watermelon Scions Grafted onto Bottle-Gourd Rootstock under Different Light Intensities

Abstract: Although grafting is broadly used in the production of crops, no information is available about the proteins involved in vascular connections between rootstock and scion. Similarly, proteome changes under the light intensities widely used for grafted seedlings are of practical use. The objective of this study was to determine the proteome of vascular connections using watermelon (Citrullus vulgaris Schrad.) ‘Sambok Honey’ and ‘Speed’ as the scion and bottle gourd (Lagenaria siceraria Stanld.) ‘RS Dongjanggun’ … Show more

“…Protein synthesis plays an important role in plant cells with regard to many physiological processes in response to unfavorable conditions [ 46 47 ]. The expression of an abundant number of protein spots under Pi starvation which plays a crucial role in protein synthesis was observed to be up-regulated under Pi starvation.…”

“…For 2D-PAGE analysis, protein extraction was carried out by following the procedure of Muneer et al [ 25 ]. In detail, leaf tissue (0.1 g) was homogenized in liquid nitrogen using a prechilled pestle and mortar.…”

“…A total of 70 μ g of dissolved protein sample was separated by 2DE in the first dimension by isoelectric focusing on a 7 cm IPG strip ( pI 4–7) (GE Healthcare, UK) and the second dimension by SDS-PAGE on a Protean II unit (Bio-Rad Hercules, USA), according to methods given by Muneer et al [ 25 ]. The samples were rehydrated for 12 h (with 125 μ L rehydration buffer containing 70 μ g proteins) before focusing.…”

“…In previous studies, the optimal quality and intensity of supplementary lighting were determined [30][31][32][33][34]. Based on these previous studies, three durations of supplementary light (8,12,16 h/day) at 100 µmol•m −2 •s −1 PPFD using mixed LEDs (W 1 R 2 B 1 ) were tested to investigate the effects of different durations of supplementary lighting on the quality of grafted watermelon plug seedlings grown in a glasshouse after the graft healing period.…”
