Proteomic Studies on Low- and High-Grade Human Brain Astrocytomas

Abstract: Human brain astrocytomas range from the indolent low-grade to the highly infiltrating and aggressive high-grade form, also known as glioblastoma multiforme. The extensive heterogeneity of astrocytic tumors complicates their pathological classification. In this study, we compared the protein pattern of low-grade fibrillary astrocytomas to that of glioblastoma multiforme by 2D electrophoresis. The level of most proteins remains unchanged between the different grade tumors and only few differences are reproducibl… Show more

“…From these reports and our former study, Prdx1 appears to have a possible correlation with an anti-tumor effect by producing epigenetic changes in cancer patients. On the other hand, some researchers have mentioned that this protein may be involved with cell proliferation and tumor growth in several kinds of solid cancers (11)(12)(13)(14)(15).…”

“…From these reports and our former study, Prdx1 appears to have a possible correlation with an anti-tumor effect by producing epigenetic changes in cancer patients. On the other hand, some researchers have mentioned that this protein may be involved with cell proliferation and tumor growth in several kinds of solid cancers (11)(12)(13)(14)(15).In order to contribute to the controversial discussion about the influence of Prdx1 expression in malignant tumors, we examined the expression of this protein in 114 cases of ESCC by immunohistochemistry and evaluated the correlations with the clinical parameters of ESCC to determine whether or not the expression of PrdxI in ESCC plays an important role in tumor progression. “…High levels expression of four proteins-VREB1, GRP78, RhoA and Rac1 and lower expression of enolase in grade IV tumors compared to lower grade tumors was also confirmed by immunohistochemistry. In another attempt to identify differentially expressed proteins between astrocytoma grades, 10 grade II and 10 grade IV samples were analyzed by 2DE followed by mass spectrometry, which resulted in the identification of 15 differentially expressed proteins (Odreman et al, 2005). These findings were subsequently validated by western blotting and immunohistochemcial staining.…”
