Proteomic profiling reveals crucial retinal protein alterations in the early phase of an experimental glaucoma model

Abstract: In conclusion, these results provide further lines of evidence that substantial molecular changes occur at the onset of the disease, identifying potential key players, which might be useful as biomarkers for diagnostics and development of medical treatment in the future.

“…IOP was elevated through thermic cauterization of episcleral veins, inducing experimental glaucoma previously shown by Anders et al 16 In parallel with the IOP elevation, the number of RGCs (839.3 6 215.4 RGC/mm 2 ) decreased by approximately 20% (***P < 0.001), compared with the contralateral controls (1070.4 6 191.2 RGC/mm 2 ) (Fig. 3).…”

“…It is expected that the wealth of information from this report will be valuable for neuroprotective ophthalmic therapy development relying on repurposing [6,8], polypharmacology [9], and target-focused new drug discovery [28], as well as for actual preventative medication aimed specifically at preserving the function of retinal ganglion cells (RGCs) from the postmenopausal impact of estrogen deficiency [29] alike. This expectation is justified especially in the context of emerging results from similar omics-driven profiling experiments on neurodegenerative diseases of the human eye [30] or animal models thereof [31][32][33][34].…”

“…Auch werden die in der RNFL befindlichen Gefäße mitberücksichtigt und können das Ergebnis verfälschen. Anders et al haben in ihren Studien allerdings die durch RNFL-Verkleinerung in OCT-Aufnahmen von Ratten angedeutete Neurodegeneration mit Auszählen der retinalen Ganglienzellen (RGC) nachgewiesen [22].…”
