Proteomic profiling of protein corona formed on the surface of nanomaterial

Abstract: The formation of protein coronas on nanomaterial will significantly alter the surface properties of nanomaterial in biological systems and subsequently impact biological responses including signaling, cellular uptake, transport, and toxicity etc. It is of critical importance to understand the formation of protein coronas on the surface of nanomaterial. Analytical techniques, especially mass spectrometry-based proteomics methods, are playing a key role for the qualitative and quantitative analyses of protein co… Show more

“…The first range of problems concerns the composition of the forming protein corona. The primary method for determination of the protein corona composition is the 'bottom-up' proteomic analysis based on the determination of masses of the peptides obtained as a result of enzymatic digestion of the corona [109,110]. In the 'shotgun' type proteomics, a composition of proteins is subjected to proteolytic digestion, and then the obtained peptides are separated using high-performance liquid chromatography (HPLC).…”

“…Elastin is the predominant protein in the extracellular matrix of arterial walls. This protein plays an important role in creating elastic properties and flexibility in the arteries when blood pressure changes, as well as in many other tissues of the body such as the lungs, skin and ligaments [ 134 , 135 ]. In their natural environment, the components of elastin first converge in the form of a water-soluble precursor molecule called their tropoelastin, and then these precursors combine to form elastin fibers by forming covalent cross-links (Fig.…”

“…MS can be used, for example, to identify (and quantify) proteins. MS has been extensively used during the last three decades to identify the proteins in the so-called “protein corona”, hence, the layer of specific proteins, from a complex biological solution, that adsorb to a specific nanoparticle [ 69 ]. MS can also be used to investigate how a protein binds to a nanoparticle by either using hydrogen/deuterium exchange [ 70 ] or limited proteolysis in combination with MS [ 51 , 71 ].…”
