Proteomic biomarkers of recovered heart function

Abstract: Aims Chronic heart failure is a costly epidemic that affects up to 2% of people in developed countries. The purpose of this study was to discover novel blood proteomic biomarker signatures of recovered heart function that could lead to more effective heart failure patient management by both primary care and specialty physicians. Methods and results The discovery cohort included 41 heart transplant patients and 20 healthy individuals. Plasma levels of 138 proteins were detected in at least 75% of these subjects… Show more

“…Preprocessing of the MRM-MS data involved several steps as described previously 21. Peptides that did not pass quality control metrics and those present in <75% of the patient samples were eliminated from the analysis.…”

“…In conclusion, the iTRAQ technology reduces the influence of system error (ion inhibition effect, background noise and equipment conditions) down to minimum and has incomparable advantages relative to other methods. The iTRAQ technology can label all proteins including phosphorylated proteins, glycosylated proteins and post-translational modifications of proteins, which provides more detailed quantitative information for the discovery and identification of multiple sample analysis and disease markers in different periods of time [7,9]. In addition, the incidence and mortality of cancer are faster growing compared with any other disease in the world.…”

“…In relation to treatment and restoring the function of the heart, plasma profiles have been determined to compare different methods in aortic surgery, to find markers of calcific aortic stenosis in patients after aortic valve replacement, and find signatures of both cardiac allograft vasculopathy and recovered heart function . Other investigations of the heart include a study of mitral valve prolapse, where Tan et al evaluated whether plasma biomarkers could be used in the evaluation of mitral regurgitation in patients.…”

“…190 Some applications in which the analysis of the plasma proteome was made in relation to transplants were mentioned in the section on circulations and cardiovascular risk. 121,191 While in such specific instances it may be difficult to find sizeable study cohorts, liver, kidney, and stem cell transplantation have also been investigated. Freue et al used LC-MALDI-TOF/TOF and iTRAQ to identify plasma proteomic signatures of allograft rejection.…”
