Proteomic analysis of synovial fluid: insight into the pathogenesis of knee osteoarthritis

Abstract: Purpose We conducted a proteomic analysis of synovial fluid (SF) to identify differentially expressed proteins and analyse their correlation with osteoarthritis (OA) severity. Our primary purpose was to gain insight into the pathogenesis of OA. Methods SF samples were acquired from 12 knee OA patients and 12 non-OA controls (ten had a meniscus injury, two had a discoid meniscus and all exhibited intact articular cartilage) and sequentially subjected to two-dimensional electrophoresis (2-DE). The radiographic g… Show more

“…The 1-year results of this subcohort are fully in line with the previously published 1-year results of the entire cohorts from both original RCTs. 21,31 One year after KJD, a statistically significant increase in medial, minimal, and mean JSW was found, this increase was still significant after 2 years. A statistically significant increase in medial and minimal JSW was found after 1 year in the HTO group, which also sustained at 2 years.…”

“…Reportedly ESWT ameliorated experimental osteoarthritic cartilage damage and alter angiogenesis pattern [15]. The preliminary proteomic data revealed abundant significant proteins that warrant further characterization [16][17][18]. These proteins of interest were reported to participate in the cellular response to physical stress, calcium homeostasis, chemotaxis and lipid oxidative stress in several tissue types under pathological contexts.…”

“…The score for the blank control group was markedly higher than for the double transfection group and single transfection groups (all p < 0.05), and the grade of double transfection group was also higher than that of the single transfection groups (p < 0.05). “…Osteoarthritis (OA) is a heterogeneous disease primarily caused by an imbalance in repair or insufficient self-repair of injured synovial joint tissue (1,2). OA affects more than 15% of the global population, imposing heavy burdens on healthcare services worldwide (1,3,4).…”
