Proteomic analysis of inviable salmonid hybrids between female masu salmon Oncorhynchus masou masou and male rainbow trout Oncorhynchus mykiss during early embryogenesis

Abstract: Early embryos of inviable hybrids between female masu salmon Oncorhynchus masou masou and male rainbow trout Oncorhynchus mykiss at 9, 12, 15 and 20 days after fertilization were examined for protein expression profiles. A total of 44 proteins, mostly down-regulated products of house-keeping genes and those involved in nucleic acid metabolism or chromatin replication, were identified in hybrid embryos by mass spectrometry analysis and protein database searching. The identified down-regulated proteins may be re… Show more

“…These included glutathione S‐transferase, UDP‐glucose 6‐dehydrogenase and glucose‐6‐phosphate 1‐dehydrogenase. Interestingly, upregulated proteins involved in the glycolytic pathway have also been reported in cancers (Masui et al ., ) and embryo development abnormality (Zheng et al ., ). Dose‐dependent downregulated proteins were apolipoprotein A‐I, vitellogenin and protein for structural maintenance of chromosomes.…”

“…After incubation at 37 °C overnight, the supernatants containing the peptides were subsequently collected after vortexing and concentrated into approximately 4 μL using a CC101 centrifugal concentrator (Tomy Inc., Tokyo, Japan). Desalting of the resulted peptides mixture, MADLI‐TOF MS analysis, and protein database searching using MASCOT server engine Version 2.2.0.7 were performed with the same method described by Zheng and others (2011). Only matched proteins with significant score ( P < 0.05) of MASCOT searches were considered positive identifications.…”
