Proteomic analysis of human serum for Finding pathogenic factors and potential biomarkers in preeclampsia

Abstract: Objective(s)-To apply a novel proteomic method to discover potential pathogenic factors and biomarkers of preeclampsia.Study design-Sera from five patients complicated with preeclampsia and five healthy pregnant controls were separately pooled. Each pool was treated with peptide ligand library beads (PLLBs) to remove high abundance proteins by affinity and thus enrich low abundance proteins. The proteins from the eluate were analyzed by a combination of 1D-Gel-LC-MS/MS. Protein expression levels were quantifie… Show more

“…Plasma levels of fi bulin-1 were identifi ed as a potential marker for kidney malfunction, 19 and increased levels of fi bulin-1 were detected in sera from patients with preeclampsia. 20 It is likely that the feature common to these diseases and the lung disease we studied is active fi brosis.…”

“…However, if plasma is pre-treated with ProteoMiner, species can be detected down to as low as 10 pg protein per mL, with an increment of sensitivity of up to three orders of magnitude. Clear advantages in accessing the low-abundance plasma proteome via CPLL treatment have also been reported by Beseme et al [31] and other groups [16,[32][33][34][35][36][37][38] who claimed the importance of early-stage biomarkers. Although after a few years of use CPLLs did not yet generate validated protein markers, a number of scientific reports place a realistic discovery/validation hope very high.…”
