Proteomic Analysis of Gastrocnemius Muscle in Rats with Streptozotocin-Induced Diabetes and Chronically Exposed to Fluoride

Abstract: Administration of high doses of fluoride (F) can alter glucose homeostasis and lead to insulin resistance (IR). This study determined the profile of protein expression in the gastrocnemius muscle of rats with streptozotocin-induced diabetes that were chronically exposed to F. Male Wistar rats (60 days old) were randomly distributed into two groups of 18 animals. In one group, diabetes was induced through the administration of streptozotocin. Each group (D-diabetic and ND-non-diabetic) was further divided into … Show more

“…Identification of peptides was performed on a nanoAcquity UPLC‐XEVO QToF mass spectometry system (Waters), as previously described (Lima Leite et al, ). Difference in expression between groups was determined using PLGS software and expressed as P < 0.05 for down‐regulated and P > 0.95 for up‐regulated proteins.…”

“…The fraction containing hydrophobic proteins was collected and the proteins were purified using detergent removal spin columns (Thermo Scientific). Proteins were extracted, reduced, alkylated, and digested as previously described, and then analyzed using a nanoAcquity UPLC‐XEVO QToF mass spectrometry system . Differences in expression between the groups were assessed using PLGS software and expressed as P < .05 for down‐regulated and (1 – P ) > 0.05 for up‐regulated proteins.…”

“…Among these are cell cycle, ion transport, apoptosis, signaling pathways and metabolism of free radicals.
