Proteomic analyses of decellularized porcine ovaries identified new matrisome proteins and spatial differences across and within ovarian compartments

Abstract: Premature ovarian insufficiency (POI) affects approximately 1% of women. We aim to understand the ovarian microenvironment, including the extracellular matrix (ECM) and associated proteins (matrisome), and its role in controlling folliculogenesis. We mapped the composition of the matrisome of porcine ovaries through the cortical compartment, where quiescent follicles reside and the medullary compartment, where the larger follicles grow and mature. To do this we sliced the ovaries, uniformly in two anatomical p… Show more

“…Therefore, the protection from the radiation injuries is of great practical significance in protecting the human body, especially for the highly metabolized and rapidly renewing germ cells. Radiation might lead to POI or even POF, the final phase of POI, which is a female endocrine disease, with the FSH ≥40 IU/L [ 18 ]. The main clinical manifestations of infertility (due to high gonadotropin and low estrogen) may cause the long-term menopausal symptoms, inducing harm to the women's physical and mental health.…”

“…There is also an opportunity to combine SW ultrasound elastography with other complementary imaging techniques, including second harmonic generation, as has been done with prostate biopsies, 66 to gain new insights into how the structure and organization of ECM proteins contribute to the mechanical properties of tissues. Finally, SW ultrasound elastography data could be used to build three-dimensional (3D) maps of SW velocity and compared to 3D maps of ECM proteins 67 to interrogate how matrix composition influences SW propagation.…”

“…56 Omics technologies such as proteomics or singlecell transcriptomics address issues with regards to characterisation of cells and matrices. [139][140][141][142] Novel non-destructive imaging techniques, such as micro-CT, optimal coherence tomography or 2photon microscopy promise to overcome the barrier of increased tissue thickness. [143][144][145] The use of second-harmonic generation or modified amino acids, such as azide-labelled proline, provides an ideal system for visualising matrix modifications.…”
