Proteome Analysis of Human Aqueous Humor

Abstract: Proteomic analysis of hAH identified 676 nonredundant proteins. More than 80% of these proteins are novel identifications. The elucidation of the aqueous proteome will establish a foundation for protein function analysis and identification of differentially expressed markers associated with diseases of the anterior segment.

“…15 TTR is present in almost all ocular tissues in noninflammatory eyes, including AH. [16][17][18] TTR in the vitreous has been demonstrated not to originate from the blood. 17 Similarly, considering its suggested production by CPE, local production of TTR in AH seems plausible.…”

“…27 In the past, AH was considered to be a plasma filtrate, 27,28 however, recent studies show the presence of many specific proteins in normal AH. 16,18 Although the functions of the majority of these proteins are not known, their presence suggests active local production by cells and tissues of the anterior segment. Expression levels of AH proteins in our study showed significant correlation with each other, which could imply their interactions in the inflammatory or, alternatively, regulatory molecular mechanisms.…”

“…It contains electrolytes, proteins, cytokines, organic solutes, and growth factors to nourish the avascular tissue of the anterior chamber. 4 Most proteins are released through active secretion. 5 Na + is released by epithelial cells of the ciliary body via membrane Na + /K + ATPases, and chloride reaches the aqueous humor via Cl -channels.…”

“…The MS scan 1 automatic gain control target was set to 1e 6 , and the MS scan 2 target was set to 5e 5 with maximum ion injection times of 50 and 100 ms, respectively. Dynamic exclusion placed selected ions on an exclusion list for 30 s (50,51). Two different preparations of the complex were independently subjected to chemical cross-linking, limited proteolysis, and MS/MS as described above, the first in the absence and the second in the presence of the reducing agent, tris(2-carboxyethyl)phosphine (50 mM final concentration) (Hampton Research).…”
