Proteoglycan degradation by the ADAMTS family of proteinases

Abstract: Proteoglycans are key components of extracellular matrices, providing structural support as well as influencing cellular behaviour in physiological and pathological processes. The diversity of proteoglycan function reported in the literature is equally matched by diversity in proteoglycan structure. Members of the ADAMTS (A Disintegrin And Metalloproteinase with ThromboSpondin motifs) family of enzymes degrade proteoglycans and thereby have the potential to alter tissue architecture and regulate cellular funct… Show more

“…Disintegrin sequences are also present in endogenous ligands such as the a disintegrin and metalloprotease (ADAM) family of metalloproteases (51). It is possible that a disintegrin may render the RGD motif inaccessible in OPN-R, and under such a circumstance, the C-ter- minal SVVYGLR may serve as an alternative cell-binding and cell-signaling motif (52,53).…”

“…[86][87][88] The family of a disintegrin and metalloproteinase with thrombospondin motifs (ADAMTS) proteinases is another group of extracellular metalloproteinases with roles in ECM degradation. 89 Their ability to cleave procollagens and proteoglycans 89,90 suggests that they may influence on cardiac ECM integrity and function during heart failure progression. Interestingly, several members of the ADAMTS family were recently shown to be upregulated in pressureoverloaded hearts, 91 however the role of this family of proteinases in the diseased heart remains to be elucidated.…”

“…It is proposed that aggrecan is degraded in the early stage of arthritis and is followed by collagen degradation. This earlier aggrecan depletion is thought to be due to the action of aggrecanases belonging to ADAMTSs family including ADAMTS-4 and -5 (Murphy and Nagase, 2008;Stanton et al, 2011). Indeed, cartilage matrix of ADAMTS-5 null mice was shown to be largely protected from challenges against antigen-induced arthritis (Stanton et al, 2005) and a mouse osteoarthritis (OA) model has been induced by surgical destabilisation of knee joints (Glasson et al, 2005).…”
