Proteins of bovine leukemia virus. I. Characterization and reactions with natural antibodies

Abstract: The bovine leukemia virus (BLV) was purified from a chronically infected fetal lamb kidney cell line. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of this virus revealed the presence of eight distinguishable viral components with molecular weights ranging from 80,000 to 11,000. The major component is a non-glycosylated protein having a molecular weight of 24,000 (p24). At least three heavier polypeptides were found, one of them representing a glycoprotein (gp 60). In addition, four minor … Show more

“…The virus was isolated from three different cell lines: (1) FLS-BLV, a BLV-producing fetal lamb spleen cell line; (2) FLK-BLV, a fetal lamb kidney cell line (Van der Maaten and Miller, 1976;Van der Maaten et al, 1974) and (3) A77th v +, a chronically infected fetal lamb thymus cell line kindly provided by Drs Onuma and Olson (Onuma et al, 1976). Virus was concentrated and purified on sucrose gradient as previously described (Deshayes et al, 1977).…”

“…Further purification of this protein is now in progress and will permit its identification. The very low level of its precipitation in RIPA (less than 5 %) could explain why no detectable reactivity was previously found using a complement-fixation test (Deshayes et al, 1977). On the other hand, it cannot be excluded from the present experiments that p12 was physically associated with either p24 or gp51.…”

“…This agent can induce lymphocyte transformation and eventually, development of lymphosarcomatous tumours (FERRER, 1980). Immunologically relevant proteins of the BLV virion include the membrane glycoproteins gp51 and gp30 and the major core protein p24 (DESHAYES et al, 1977(DESHAYES et al, , 1980. Several reports have shown that BLV-infection is widespread in cattle from the American continent ( M A R~N et al, 1978;SAMAGH and KELLAR, 1982; DIGIACOMO, 1992).…”
