Proteins iodinated by the Chloramine-T method appear to be degraded at an abnormally rapid rate after endocytosis

Abstract: Proteins iodinated by the chloramine-T method appear to be degraded at an abnormally rapid rate after endocytosis ( Proteins labeled with either 3H by reductive methylation or 125I by the chloramine-T method were incubated with Xenopus laevis oocytes; the incorporation and acid precipitability of the proteins were then studied. The uptake rates of both specifically incorporated (vitellogenin) and nonspecifically incorporated proteins (bovine serum albumin and X.

“…6, B and C) and the absence of significant degradation of VTG during its incorporation by oocytes (18) suggest that the light platelets are not simply a degradative compartment entered by a small percentage of the total ligand. The reason(s) for the second increase in density of yolk platelets to 1.23 g/cc are not currently known, nor is the mechanism for the apparent loss in enzyme activity during this final transition.…”

“…But iodine radioisotope, which was labeled by the above method, was normally found to be unstable in living subjects and such deiodination resulted in high background or false signals. [14][15][16] Moreover direct radiolabeling using a strong oxidant for the formation of reactive iodo species often cause decreased biological activity of the substrate. Therefore several prosthetic groups have been developed for indirect labeling of iodine radioisotopes.…”

“…24 Radioactive counting was done with a γ-ray spectrometer after separating the melatonin from the reaction mixtures. 125 I]-Iodomelatonin administration, the rats were sacrificed through cervical dislocation and the amount of radioactivity in each tissue was determined with a γ-ray spectrometer and expressed as % uptake of total amount of injected dose per g of tissue.…”
