Protein-protein interactions in the yeastPKC1 pathway: Pkc1p interacts with a component of the MAP kinase cascade

Abstract: The two-hybrid system for the identification of protein-protein interactions was used to screen for proteins that interact in vivo with the Saccharomyces cerevisiae Pkc1 protein, a homolog of mammalian protein kinase C. Four positive clones were isolated that encoded portions of the protein kinase Mkk1, which acts downstream of Pkc1p in the PKC1-mediated signalling pathway. Subsequently, Pkc1p and the other PKC1 pathway components encoding members of a MAP kinase cascade, Bck1p (a MEKK), Mkk1p, Mkk2p (two func… Show more

“…Interestingly, however, two-hybrid analysis indicates that Pkc1p interacts with Mkk1p but not with the immediate downstream kinase, Bck1p. Mkk1p also interacts with Bck1p and Mkk2p, and Mkk2p interacts with Slt2/Mpk1p (450,567). Taken together, these observations indicate that, as in the HOG pathway, the MAPKK could play an important role in mediating the formation of a complex between pathway components.…”

“…Mutation of Thr1119 to Ala, Cys, or Tyr also results in constitutive signaling (D. E. Levin and A. K. Sobering, unpublished), suggesting that disruption of an interaction involving Thr1119 (either by phosphorylation or mutation) is the key to activation of this MEKK. Bck1 is presumed to phosphorylate and activate Mkk1/2 based on genetic epistasis studies, two-hybrid interaction, and its requirement for activation of Mpk1 (137,148,254). Mkk1/2 phosphorylate Mpk1 on neighboring tyrosyl and threonyl residues in a T-X-Y motif that is diagnostic for MAP kinases.…”

“…Pkc1 phosphorylates Bck1 in vitro at several sites in a hinge region between its putative regulatory domain and its catalytic domain Levin 2005) that is also the site of activating mutations (Lee and Levin 1992). Bck1 is presumed to phosphorylate and activate Mkk1/2 on the basis of genetic epistasis studies, two-hybrid interactions, and its requirement for activation of Mpk1 Kamada et al 1995;Paravicini and Friedli 1996;Ho et al 2002). Mkk1 and -2 phosphorylate Mpk1 on neighboring tyrosyl and threonyl residues in a T-X-Y motif within the activation loop conserved among MAPKs.…”
