“…Later work discovered that the ortho -quinone functional group generated upon tyrosine oxidation undergoes a reaction with the sulfhydryl group of a free cysteine residue. , This approach was used to prepare conjugates of sfGFP with three different proteins: CRISPR-Cas9, a HER2-binding scFv, and nanoluciferase . Further work explored the use of different tyrosinases to expand the scope of tyrosine residues that could be targeted in this manner .…”

“…Examples of this include the attachment of a drug for drug delivery, [6][7][8][9] a fluorophore for visualization [10,11] or a polyethylene glycol (PEG) to improve pharmacokinetics and pharmacodynamics of a protein therapeutic. [12][13][14] Advanced constructs, where a protein is conjugated to another protein [15,16] or oligonucleotides, [17][18][19] have also emerged. This demonstrates the opportunities selective protein conjugation strategies can provide.…”

“…The applications of quinone oxidation have been largely limited to small molecule transformations despite the significant research progress in this area 17,18 . On the other side, in nature, the reactions between oquinones such as DOPA 19 and protein side chains are important but complex in fundamental process of life 20 . Although the early work provided by Mason in 1955 showed that o-quinones could react with proteins via the N-terminal residue 21 , the reaction between o-quinone motifs and proteins have not been fully elucidated on a molecular level 22 .…”
