Protein phosphatase type 2C PP2CA together with ABI1 inhibits SnRK2.4 activity and regulates plant responses to salinity

Abstract: Protein phosphatases 2C (PP2Cs) are important regulators of plant responses to abiotic stress. It is established that clade A PP2Cs inhibit ABA-activated SNF1-related protein kinases 2 (SnRK2s). Our recently published results show that ABI1, a member of clade A of PP2C is also a negative regulator of SnRK2.4, a kinase not activated in response to ABA. Here, we show that another member of this clade - PP2CA, interacts with and inhibits SnRK2.4. The salt-induced SnRK2.4/SnRK2.10 activity is higher in abi1–2 pp2c… Show more

“…SnRK2.4 and OST1 were comparably inhibited by the ABI1‐related subclass of ABA coreceptors including ABI2, HAB1, and HAB2 but SnRK2.4 was less efficiently regulated by PP2CA‐type phosphatases encompassing PP2CA, HAI1 to HAI3, and AHG1. The finding is consistent with the reported inhibition of SnRK2.4 by ABI1 and PP2CA (Krzywińska et al , ).…”

“…Overexpression of some PYL, PP2C, and SAPK2 genes significantly increases or decreases drought tolerance in transgenic plants [ 56 58 ]. Moreover, the ABA receptor PYL5 can inhibit the activity of clade A PP2Cs [ 59 ], and PP2CA together with ABI1 inhibits SnRK2.4 activity and regulates plant responses to salinity [ 60 ]. These data clarify the function and nature of the cascade relationship of these genes in the stress response of plants.…”

“…Salt stress also triggers abscisic acid (ABA) pathways and osmotic stress signaling. Salt treatment increases ABA concentrations in plant cells (Duan et al ., ; Geilfus et al ., ) and activates sucrose nonfermenting 1‐related protein kinase 2 (SnRK2) kinase activity (Krzywińska et al ., ), and salt treatment of a protein phosphatase 2c ( pp2c ) double mutant ( abi1‐2 pp2ca‐1 ) leads to higher SnRK2 kinase activity (Krzywińska et al ., ). However, some evidence also indicates that salt/osmotic stress‐mediated SnRK2 activation is independent of ABA signaling (Boudsocq et al ., ; Zhang H. et al ., 2011; Zhang et al ., ).…”
