Protein Levels of Several Arabidopsis Auxin Response Factors Are Regulated by Multiple Factors and ABA Promotes ARF6 Protein Ubiquitination

Abstract: The auxin response factor (ARF) transcription factors are a key component in auxin signaling and play diverse functions in plant growth, development, and stress response. ARFs are regulated at the transcript level and posttranslationally by protein modifications. However, relatively little is known regarding the control of ARF protein levels. We expressed five different ARFs with an HA (hemagglutinin) tag and observed that their protein levels under the same promoter varied considerably. Interestingly, their p… Show more

“…ARF7 and ARF19 are two class-A ARFs that are both regulated by the proteasome and directly interact with AFF1. Similarly, the class-A ARF6 13 , 14 and ARF8 13 , the Class-B ARF1 15 , ARF2 16 , and the Class-C ARF17 13 also undergo proteasome-dependent degradation. Whether these protein stabilities are regulated through the activity of SCF AFF1 or through another mechanism remains unknown.…”

“…Finally, the clearest example of ARF ubiquitination has been described for AtARF6. The mechanism is induced by abscisic acid and leads to the degradation of the protein (Li et al ., 2020).…”

“…The HA-cyFBP protein was pulled down from the total protein extract as described [ 70 ]. The anti-HA agarose beads (Sigma-Aldrich) were added into the total protein extract (1 mg of total proteins) and incubated at 4 °C for 1 h (on a tumbler).…”

“…The bound proteins were eluted with 1 mg ml −1 HA peptide (GenScript, Piscataway, USA). After adding the 4X SDS loading buffer and heat treatment at 95 °C for 5 min, the samples were then analyzed by electrophoresis and Western blotting for HA-cyFBP and ubiquitinated HA-cyFBP as described [ 70 ].…”
