Protein interaction domains of the measles virus nucleocapsid protein (NP)

Abstract: The currently accepted model for measles virus (MV) transcription and replication assumes the nucleocapsid (NP) protein to possess the ability to bind to RNA, to other NP molecules, and to the phosphoprotein (P) during ribonucleocapsid (RNP) assembly, as well as to the matrix protein (M) during virion assembly. We have cloned the MV NP open reading frame and have expressed the protein in bacteria as a fusion with glutathione-S-transferase (GST). Affinity purified GST-NP fusion protein has been used as a probe … Show more

“…We have shown that enhanced sensitivity to 1,25-(OH) 2 D 3 is an intrinsic property of OCL precursors from Paget's patients, and that it may result from increased affinity of the vitamin D receptor for 1,25-(OH) 2 D 3 (26). Similarly, OCL precursors transduced with the MVNP gene showed increased expression of 24-hydroxylase mRNA in response to 1,25-(OH) 2 (27) demonstrated that MVNP proteins can interact with a variety of cellular proteins. Immunoprecipitation studies using glutathione S-transferase nucleocapsid protein constructs demonstrated that MVNP interacted specifically with at least 12 cellular proteins.…”

“…As discussed above, MV N TAIL is required for stable binding of N to P (18,20,26). To determine whether the isolated N TAIL domain was able to bind P, we tested the ability of bacterial lysates expressing hexahistidine tagged N TAIL to pull down untagged P onto an IMAC resin.…”

“…In Morbilliviruses and Respiroviruses, the region responsible for binding of nucleocapsids to P is located within N TAIL (13,18,20,24). Consequently, N TAIL is required for N⅐RNA to act as a template for viral RNA synthe-sis (13,16,25).…”
