Protein-Based Drug Delivery Nanomedicine Platforms: Recent Developments

Abstract: Background: Naturally occurring protein cages, both viral and non-viral assemblies, have been developed for various pharmaceutical applications. Protein cages are ideal platforms as they are compatible, biodegradable, bioavailable, and amenable to chemical and genetic modification to impart new functionalities for selective targeting or tracking of proteins. The ferritin/apoferritin protein cage, plant-derived viral capsids, the small Heat shock protein, albumin, soy and whey protein, collagen, and gelatin hav… Show more

“…Because of these residues, gelatin often exhibits amphoteric behavior, demonstrating different physicochemical properties at different pH values ( Stagnoli et al, 2023 ). The amphoteric behavior characteristics of gelatin have been developed to deliver various drugs to treat cancer ( Aljabali et al, 2022 ). Another characteristic of gelatin is viscosity, which increases with polymer concentration and decreases with temperature and pH ( Milano et al, 2023 ).…”

“…And Figure 3 represents the nano-delivery technologies for mRNA vaccines. The latter mainly consists of protein cages such as ferritin and albumin cages ( Aljabali et al, 2022 ), polymer and lipid groups. And polymers could be classified according to whether their components are synthetic or natural ( Yan and Zhou, 2023 ).…”

“…The use of proteins as suitable biomaterials for pharmaceutical applications is a well-known strategy to modulate the physico-chemical and biopharmaceutical features of the active compounds ( Martínez-López et al, 2020 ; Can Karaca et al, 2023 ). The specific structural properties of proteins can promote the development of various formulations such as micro/nanoparticles, scaffolds, (hydro)gels and films, offering a wide range of administration routes for the entrapped compound(s) ( Elzoghby et al, 2012 ; Aljabali et al, 2022 ). A milestone of the use of protein for drug delivery purposes is represented by human albumin exploited for the development of nanoparticles containing paclitaxel ( Spada et al, 2021 ).…”
