Protein Assembly by Orthogonal Chemical Ligation Methods

Abstract: Chemical synthesis harbors the potential to provide ready access to natural proteins as well as to create nonnatural ones. The Staudinger ligation of a peptide containing a C-terminal phosphinothioester with a peptide containing an N-terminal azide gives an amide with no residual atoms. This method for amide bond formation is orthogonal and complementary to other ligation methods. Herein, we describe the first use of the Staudinger ligation to couple peptides on a solid support. The fragment thus produced is u… Show more

“…[15,16] In an impressive effort demonstrating the power of methods involving orthogonal chemical ligation, Raines and co-workers completed the total synthesis of ribonuclease A (31, RNAse A, 124 amino acids) by linking three fragments (Scheme 7). [17] RNAse A(110-111) was synthesized as the Cterminal phosphanyl thioester FmocCys(Trt)Glu(OtBu)- …”

“…20 The initial intermediate is an iminophosphorane that rearranges to an amido phosphonium salt which hydrolyses to the amide (Scheme 8). Azides are involved in the synthesis of proteins by native chemical ligation (NCL), the method of choice for the preparation of large peptides and proteins (see later).…”
