Objectives A large amount of fructose is metabolized in the liver and causes hepatic functional damage. Δ9‐tetrahydrocannabinol (THC) is known as a therapeutic agent for clinical and experimental applications. The study aims to investigate the effects of THC treatment on inflammation, lipid profiles and oxidative stress in rat liver with hyperinsulinemia. Methods Sprague‐Dawley rats were divided into groups: control, fructose (10% fructose in drinking water for 12 weeks), THC (1.5 mg/kg/day for the last 4 weeks, intraperitoneally) and fructose+THC groups. Biochemical parameters were measured spectrophotometrically. ELISA method was used for insulin measurement. Apoptosis and inflammation markers were detected by the streptavidin‐biotin peroxidase method. Key findings The consumptions of food and fluid are inversely proportional to fructose and non‐fructose groups. Insulin levels were the highest in fructose group. The reduced glutathione‐S‐transferase level significantly increased in fructose + THC group compared with fructose group. Total cholesterol level in the fructose + THC group was higher than the fructose group. Caspase‐3 and NF‐κβ immunopositive cell numbers increased in fructose + THC rats compared with fructose group. The number of IL‐6 immunopositive cell decreased in fructose + THC group compared with fructose group. Conclusions According to the result, long‐term and low‐dose THC administration may reduce hyperinsulinemia and inflammation in rats to some extent.

Association of micronuclei frequency and other nuclear anomalies with flaxseed diet in metabolic syndrome patients

Introduction: This study aimed to investigate whether a rich flaxseed diet could modify the micronuclei frequency, nuclear anomalies and biochemical and anthropometric parameters in metabolic syndrome patients. Material and methods: Fifty-eight patients with metabolic syndrome were dichotomized into two groups, namely, the control group (CG) and the intervention group (IG), and the patients in the IG received a diet supplemented with flaxseed. The anthropometric and clinical parameters were measured. Nuclear anomalies isolated from the oral mucosa were analyzed at the beginning and end of the 3-month follow-up. Results: After the nutritional follow-up, the percentage of fat was decreased in the CG (before, 45.40%; after, 37.89%, p < 0.001). In the IG, the weight (before, 81.03 ±13.17 kg; after, 79.81 ±13.47 kg, p = 0.01), body mass index (before, 32.7 ±5.4; after, 32.2 ±5.6, p = 0.009) and very-low-density lipoprotein (before, 31.96 ±15.23; after, 26.13 ±7.54, p = 0.04) were decreased. The frequencies of MN (micronucleated) and BN (binucleated) cells were decreased in the CG (before, 14.03 ±5.28, after 11.17 ±3.68; before, 2.66 ±3.5, after, 1.10 ±1.66; p = 0.001/0.02, respectively), while in the IG, the frequencies of MN and BN cells and NB (nuclear buds) were decreased (MN before, 4.97 ±5.

Secoisolariciresinol diglucoside (SDG) is the main component of lignans with various biological activities, including anticancer activity. However, whether SDG has obvious anticancer effects on colorectal cancer (CRC) is unclear. Pyroptosis, a form of programmed cell death, has received increasing attention in cancer-related research. In this study, we aimed to test the anticancer properties and relatecd functional mechanisms of SDG. we found that SDG not only inhibited the cell viability of HCT116 cells, but also induced HCT116 cells to swell with apparent large bubbles, which are typical signs of pyroptosis. Furthermore, SDG induced cell pyroptosis by enhancing cleavage of the N-terminal fragment of gasdermin D (GSDMD) in CRC cells, accompanied by increased caspase-1 cleavage. Consistent with this, SDG-induced GSDMD-N-terminal fragment cleavage and pyroptosis were reduced by siRNA-mediated silencing of caspase-1 or treatment with the specific caspase-1 inhibitor VX-765 treatment, suggesting that active caspase-1 further induces pyroptosis. A mechanistic study showed that SDG induced reactive oxygen species (ROS) accumulation and inhibits phosphatidylinositol 3-kinase (PI3K) phosphorylation and increases pyroptosis, while increasing GSDMD and caspase-1 cleavage and enhancing expression of BCL2-associated X (BAX), which could be rescued by the ROS scavenger (NAC), suggesting that SDG-induced GSDMEdependent pyroptosis is related to the ROS/PI3K/AKT/BAX-mitochondrial apoptotic pathway. In vivo results showed that SDG significantly inhibited tumor growth and induced pyroptosis in the HCT116-CRC nude mouse model. In conclusion, our findings suggest that the anticancer activity of SDG in CRC is associated with the induction of GSDMD-dependent pyroptosis by SDG through the generation of ROS/ P13K/AKT/BAK-mitochondrail apoptosis pathway, providing insights into SDG in its potential new application in cancer treatment.
