Protective effects of the novel adenosine derivative WS0701 in a mouse model of posttraumatic stress disorder

Abstract: Aim: To investigate the effects of the novel N6-substituted adenosine derivative {(2R,3S,4R,5R)-3,4-dihydroxy-5- [6-[(4-hydroxy-3-methoxybenzyl)amino]-9H-purin-9-yl]tetrahydrofuran-2-yl} methyl decanoate (WS0701) on stress-induced excessive fear, anxiety, and cognitive deficits in a mouse model of posttraumatic stress disorder (PTSD). Methods: Male mice underwent a conditioned foot shock and single prolonged stress procedure to induce PTSD. Contextual/cued fear, elevated plus-maze, open field and novel object … Show more

“…However, the interpretation of these results is confounded by re-exposure to water, which is a reminder of the stressor in SPS-exposed rats but not controls ( 106 ) and so may affect performance in this task. We and others have also observed impairment in object recognition ( 148 , 149 ) and social discrimination ( 148 ). This impairment in social discrimination may be attributed to decreased behavioral flexibility, a notion corroborated by evidence that SPS decreases performance in set shifting tasks ( 150 – 152 ).…”

“…A 2A R mediated increased interleukin (IL)-1β in the brain contributed to anxiety ( Chiu et al, 2014 ). Fear and anxiety in a mouse model of post-traumatic stress disorder were alleviated by administration of a derivative of adenosine, WS0701 ( Huang et al, 2014 ). A 1 R agonists have been used to treat anxiety, but have troublesome side-effects so positive allosteric modulators have been developed as potent anxiolytic agents ( Vincenzi et al, 2016 ).…”

“…Adenosine, via A2A receptors, increased interleukin 1 in the brain contributing to anxiety [118]. Administration of a derivative of adenosine, WS0701, alleviated anxiety and fear in a mouse model of posttraumatic stress disorder [119]. In the aetiology of anxiety disorders and addiction, the basolateral amygdala plays a critical role and adenosine, via activation of A2A receptors, facilitated basolateral amygdala pyramidal cell output [120].…”
