Protective effects of resveratrol on postmenopausal osteoporosis: regulation of SIRT1-NF-κB signaling pathway

Abstract: Postmenopausal osteoporosis severely jeopardizes human health. Seeking for therapeutic drugs without side effects is of great necessity. Our study was designed to investigate whether resveratrol, an agonist of SIRT1, could have favorable effect on osteoporosis and to explore the underlying mechanisms. Rat osteoporosis model (ovariectomy group, OVX) was established by bilateral ovariectomy. respectively; P < 0.05). Serum markers alkaline phosphatase (ALP) and osteocalcin were moderately restored by resveratrol.… Show more

“…Additionally, Feng et al suggested that there is a reverse correlation between BMD and OC levels in osteoporotic rat models, i.e., BMD values decreased in parallel with increased OC serum levels [48]. These results show that the levels of osteocalcin show the direct synthesis and activity of osteoblasts.…”

“…The role of another cysteine protease inhibitor, CST3, was reported to reduce the formation of osteoclasts by interfering with the late differentiation of osteoclast precursors . Studies have shown that inhibition of the NF‐κB pathway could relieve OP . Moreover, cystatin was demonstrated to affect the NF‐κB pathway .…”

“…Additionally, the rate of bone resorption exceeds the rate of bone formation of osteoclasts largely contributes to OP in postmenopausal women . The NF‐κB pathway could mediate osteoblast differentiation, and inhibition of the NF‐κB pathway promoted osteoblast differentiation and suppressed osteoclastogenesis . In the present study, we investigated the effect of CST5 on the activation and bone resorption of osteoclasts through regulation of the NF‐κB pathway, thus affecting the progression of OP.…”

“…Recently, a study by Feng et al tested the ability of resveratrol, a plant polyphenol acting as an agonist of Sirt1, to promote osteogenesis in vitro and prevent bone loss in animal models and, thus, its possible protective effect on osteoporosis [28]. In rats with bilateral ovariectomy, resveratrol moderately restored the serum level of ALP and OCN and improved bone structure.…”
