<p>The current study was designed to determine the effects of dietary quercetin on epididymal sperm and testicular changes in male rabbits during summer heat stress (HS). Twelve adult male New Zealand white rabbits were submitted to summer heat conditions (temperature-humidity index 29.9±1.2). One group was fed a basal ration (BR; n = 6), whereas the other group was fed the same basal diet supplemented with quercetin (30 mg/kg d; Que-BR; n = 6) for 60 d, and both groups were sacrificed at the end of the experiment. Epididymis and testicles isolation was done for sperm, histopathology and apoptosis assessments. The results showed that quercetin improved epididymis weight, but did not affect other testicular dimensions except testicular length. A significant improvement was observed in epididymal sperm motility, concentration, kinematic parameters, viability, mitochondrial potential and acrosome integrity in Que-BR compared to the BR group. Lowered serum malondialdehyde level was observed in quercetin supplemented rabbits. Moreover, the quercetin supplementation maintained the interstitial stroma, seminiferous tubules architecture, germinal and Sertoli cells under HS, decreasing the apoptotic germ cell rate in seminiferous tubules. In conclusion, HS condition affects the sperm and testes configurations in rabbits and dietary quercetin minimises oxidative stress, which in turn protects the testes and sperm against HS induced damage.</p>

Cadmium (Cd) damages the thyroid gland. We evaluated the effects of myo-inositol (MI), seleno-L-methionine (Se) or their combination on the thyroids of mice simultaneously administered with Cd chloride (CdCl2). Eighty-four male mice were divided into 12 groups (seven mice each). Six groups (controls) were treated with 0.9% NaCl (vehicle), Se (0.2 mg/kg/day), Se (0.4 mg/kg/day), MI (360 mg/kg/day), MI+Se (0.2 mg/kg) and MI+Se (0.4 mg/kg). The other six groups were treated with CdCl2 (2 mg/kg), CdCl2+MI, CdCl2+Se (0.2 mg/kg), CdCl2+Se (0.4 mg/kg), CdCl2+MI+Se (0.2 mg/kg) and CdCl2+MI+Se (0.4 mg/kg). An additional group of CdCl2-challenged animals (n = 7) was treated with resveratrol (20 mg/kg), an effective and potent antioxidant. All treatments lasted 14 days. After sacrifice, the thyroids were evaluated histologically and immunohistochemically. CdCl2 reduced the follicular area, increased the epithelial height, stroma, and cells expressing monocyte chemoattractant protein-1 (MCP-1) and C-X-C motif chemokine 10 (CXCL10). CdCl2+Se at 0.2/0.4 mg/kg insignificantly reversed the follicular and stromal structure, and significantly decreased the number of MCP-1 and CXCL10-positive cells. CdCl2+MI significantly reversed the thyroid structure and further decreased the number of MCP-1 and CXCL10-positive cells. CdCl2+MI+Se, at both doses, brought all indices to those of CdCl2-untreated mice. MI, particularly in association with Se, defends mice from Cd-induced damage. The efficacy of this combination was greater than that of resveratrol, at least when using the follicular structure as a read-out for a comparison. We suggest that the use of these nutraceuticals, more specifically the combination of MI plus SE, can protect the thyroid of Cd-exposed subjects.

Acrylamide (AA) is a well-known potent carcinogen and neurotoxin that has been recently linked to atherosclerotic pathogenesis. The present study is aimed at investigating the protective effect of silymarin (SIL) as an antioxidant against AA-induced hyperlipidemic cardiomyopathy in male rats. The obtained results showed that animals exposed to AA exhibited a significant increase in the levels of cardiac serum markers, serum total cholesterol, triglycerides, low-density lipoprotein cholesterol, and very-low-density lipoprotein cholesterol with a significant decrease in high-density lipoprotein cholesterol. Furthermore, AA intoxication significantly increased the malondialdehyde level (a hallmark of lipid peroxidation) and reduced antioxidant enzyme activities (i.e., superoxide dismutase, catalase, and glutathione peroxidase). SIL administration significantly attenuated all these biochemical perturbations in AA-treated rats, except for the decreased high-density lipoprotein cholesterol. Our results were confirmed by histopathological assessment of the myocardium. In conclusion, this study demonstrated a beneficial effect of SIL therapy in the prevention of AA-induced cardiotoxicity by reversing the redox stress and dyslipidemia in experimental animals.
