Protective Effects of Oral Microencapsulated Mycoplasma hyopneumoniae Vaccine Prepared by Co-Spray Drying Method.

Abstract: ABSTRACT. The efficacy of Mycoplasma hyopneumoniae oral vaccine was investigated in microsphere dosage form. A co-spray drying process was used to apply an encapsulating material, Eudragit L30 D-55, to microspheres containing Mycoplasma hyopneumoniae antigens. The microspheres were generally effective (>93%) with protein release at pH 7.4, but almost none were released at pH 1.2, for 3 hr in an in vitro dissolution test. An SPF-swine model was used to evaluate the effectiveness of the microspheres as an oral v… Show more

“…Investigation of new vaccines is actively occurring, including the use of aerosol and feed-based vaccines as well as subunit and DNA vaccines (Fagan et al, 2001;Lin et al, 2003;Murphy et al, 1993). Intradermal vaccination with a commercial bacterin has been shown to be efficacious (Jones et al, 2004).…”

“…Another possible reason for this failure is that these vaccines were based on either whole inactivated MmmSC or the cytotoxic LppQ protein. In contrast, oral delivery in pigs of another pathogenic mycoplasma, Mycoplasma hyopneumoniae, included in microspheres led to effective protection [42]. Therefore, these new antigen delivery systems might offer promising approaches for mucosal vaccine delivery in large animals.…”

“…Antigens from Mycoplasma hyopneumoniae and Actinobacillus pleuropneumoniae coated with acrylic resins (cellulose acetate phthalate and methacrylic acid) to protect them from the low pH of the stomach and released in intestines have been tested and shown efficacy when given orally to swine [89,90]. Fimbriae of ETEC in a cellulose-based delivery system reduced disease in experimentally infected pigs [150].…”
