Protective effects of adiponectin against renal ischemia‐reperfusion injury via prostacyclin‐PPARα‐Heme oxygenase‐1 signaling pathway

Abstract: Adiponectin (APN), a circulating adipose-derived hormone that regulates inflammation and energy metabolism, has beneficial effects on the cardiovascular disorders. Serum APN levels are lower in patients with coronary artery disease and higher in patients with chronic kidney disease. However, the precise role of APN in acute reno-vascular disease is not clear. Results of the present study show that serum APN concentration decreased after renal ischemia reperfusion (I/R) injury in mice. In addition, I/R-induced … Show more

“…Because antiapoptotic effects of adiponectin may result from its antiinflammatory or antioxidative effect after I/R primarily through the ADIPOR2-proliferator-activated receptor a-heme oxygenase-1 pathway, 19 inhibiting ADIPOR2 by miR-494 would result in greater inflammation and hence, more damage to the kidneys. With the relationship between IGF1R and the kidneys, IGF1R has been implicated in normal mammalian glomerular integrity, 20 and suppression of IGF1R by miR-494 may inhibit podocyte cell outgrowth, leading to proteinuria.…”

“…The above-mentioned studies should be considered separately from studies investigating the role of adiponectin in the setting of acute renal injury. The latter group of studies includes two trials investigating whether adiponectin can modulate the adverse effects of renal ischemia-reperfusion injury in mice (Cheng et al 2012, Jin et al 2013. These studies yielded conflicting results with regard to the acute responses of various parameters of renal structure and function resulting from acute renal injury.…”

“…We showed that LA-induced HO-1 expression was dependent on PPARα, whereas SAM induced HO-1 in a PPAR-independent manner. PPARα is a transcription factor responsible for fatty acid oxidation, and the PPARα/HO-1 signaling pathways constitute a protective mechanism against various cellular insults (Cheng et al, 2012;Yu et al, 2010). The precise mechanism responsible for the difference observed in HO-1 induction by LA and SAM awaits future studies.…”
