Ischemic acute renal failure is a condition that extends subsequent to sudden and momentary fall in overall or regional blood flow to the kidney. The present investigation was deliberated to scrutinize the renoprotective potential of berberine in animal model of renal ischemia reperfusion (RIR) induced dent via assessment of various biochemical and molecular biomarkers. Male Wistar rats were anesthetized and the right kidney was removed through a small flank incision. Renal ischemia reperfusion was persuaded in uni-nephrectomized rats by occlusion of left renal artery for 45 min and reperfusion for 4 weeks. After 4 weeks of treatment of berberine (10, 20, and 40 mg/kg, p.o.), hemodynamic and left ventricular function were evaluated. Induction of ischemia reperfusion resulted callous mutilation in kidney which was confirmed by alterations in oxidative stress (SOD, GSH, and MDA), membrane bound enzymes, kidney function markers (serum creatinine and BUN), and mitochondrial dysfunction. Moreover, RIR injury exhibited incredible alterations in mRNA expression of KIM-1, NGAL, Caspase-3, Bax, Bcl-2, and TNF-a levels. Conversely treatment of berberine (20 and 40 mg/kg) significantly (p50.01 and p50.001) restored ischemia reperfusion induced marring via intonation of biochemical and molecular biomarkers. To sum up, berberine demonstrated compelling renoprotective effect in RIR injury via caspase-mitochondria-dependent pathway.

There is little information about the hepatoprotective effects of gallic acid against ischemiareperfusion (I/R) damage. Animals were subjected to I/R. Gallic acid at doses of 50 and 100 mg/kg body weight (bw) were injected as a single dose prior to ischemia. Liver tissue homogenates were used for the measurement of malondialdehyde (MDA), catalase (CAT) and glutathione peroxidase (GPx) levels. At the same time alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) were assayed in serum samples and compared statistically. While the ALT, AST, LDH activities and MDA levels were significantly increased, CAT and GPx activities significantly decreased in only I/Rinduced control rats compared to normal control rats (P \ 0.05). Treatment with gallic acid at a dose of 100 mg/kg bw significantly decreased the ALT, AST, LDH activities and MDA levels, and markedly increased activities of CAT and GPx in tissue homogenates compared to I/R-induced rats with no treatment group (P \ 0.05). In oxidative stress generated by hepatic ischemia-reperfusion, gallic acid contributes partially an alteration in the delicate balance between the scavenging capacity of antioxidant defense systems and free radicals in favour of the antioxidant defense systems in the body.

Thirty-five Sprague-Dawley male rats were randomly separated into five groups comprising control, rutin 150 mg, OA, rutin 75 mg + OA and rutin 150 mg + OA. In the rutin 75 mg + OA and rutin 150 mg + OA groups, the lung malondialdehyde level (MDA) was significantly lower than that of the OA group. In the rutin 75 mg + OA and and rutin 150 mg + OA groups, the lung GPx (glutathione peroxidase), CAT (catalase) and SOD (superoxide dismutase) activities and GSH (glutathione) levels were significantly higher than those of the OA group, and significantly lower than those of the control group. iNOS expressions in the interstitial parts of the lungs were significantly lower than those of the OA group. The iNOS expression was lower in the 150 mg + OA group compared to the rutin 75 mg + OA group. It was concluded that on the ALI induced by OA, rutin had protective effects through the antioxidant and antiinflammatory properties and that the treatment of rutin as a supportive treatment in ALI was found to be practically useful. Rutin 75 mg + OA ve rutin 150 mg + OA gruplarında akciğer malondialdehit (MDA) düzeyi OA grubununkine göre önemli düzeyde düşüktü. Rutin 75 mg + OA ve rutin 150 mg + OA gruplarında akciğer GPx (glutasyon peroksidaz), CAT (katalaz), SOD (süperoksit dismutaz) aktiviteleri ile GSH (glutatyon) düzeyleri OA grubununkilere göre önemli düzeyde yüksekti ve kontrol grubununkilere göre önemli düzeyde düşüktü. Rutin 75 mg + OA ve rutin 150 mg + OA gruplarında akciğerlerin interstisyel kısımlarında iNOS ekspresyonları OA grubunkilere göre önemli düzeyde düşüktü. OA ile oluşturulan ALI'de rutinin antioksidan ve antiinflamatuvar özellikler aracılığıyla koruyucu etkilere sahip olduğu, ALI'de destekleyici tedavi amacıyla rutin uygulamasının pratik olarak yararlı olduğu tespit edildi.
