Till now, the exact role of melatonin in male infertility hasn't been fully discovered. Moreover, the intracellular signaling pathways activated by melatonin in buffalo spermatozoa especially on the level of cryopreservation process haven't been reported. The current study aimed simply to clarify the precise role of this hormone during buffalo semen cryopreservation. Semen samples were obtained randomly from 6 fertile buffalo bulls (aged 3 to 5 years). Weekly, two consecutive ejaculates were collected from each bull for successive six weeks duration using an artificial vagina. The ejaculates were pooled to eliminate variability between the evaluated samples. After that, semen samples were subjected to extension with Tris-based extender supplemented with different concentrations of melatonin (0.1, 0.25, 0.5, 0.75 and 1 mM) Vs. Trisbased extender only (control). Then they were processed to cryopreservation and thawing to assess the different semen characteristics. It had been found that 0.1 mM of melatonin significantly (p<0.05) improved post thawing motility, viability index, acrosomal integrity, total antioxidant capacity, superoxide dismutase and glutathione reductase activity. On the other hand, it significantly (p<0.05) decreased the rate of lipid peroxidation, aspartate amino transferase, alanine aminotransferase, alkaline phosphatase and DNA fragmentation. All of these previously enhanced semen characteristics were reflected positively on its in vitro fertilizing capacity, as well as the percent of harvested embryos. In conclusion melatonin supplementation to the extension media of buffalo semen during processing significantly enhanced its characteristics. Moreover, the preceding results focusing more light on the potential roles of melatonin in regulating male reproduction.

Background and Objective. Nonylphenol (NP), as an environmental pollutant, has adverse effects on the reproductive and nervous systems through disrupting the endocrine system and inducing oxidative stress. This study was aimed at investigating the protective effect of melatonin (MLT) on NP-induced reproductive and behavioral disorders in first-generation adult male rats. Methods. Pregnant Wistar rats were randomly assigned into five groups: control, NP (25 mg/kg), NP (25 mg/kg)+MLT (10 mg/kg), NP (25 mg/kg)+MLT (20 mg/kg), and MLT (20 mg/kg). The duration of treatment was 21 days from gestation time. Morris water maze was used to assess learning and memory. NP concentrations of serum and testicular tissue were measured by HPLC. Histological analysis of testicular tissues was done by H&E staining. Results. Behavioral study showed that NP does not impair learning and memory in first-generation rats. Histomorphometric results showed that NP can significantly reduce the cross-sectional area of the seminiferous tubules and the epithelium, the diameter and number of seminiferous tubules, the thickness of the epithelium, and the number of spermatocytes and spermatogonia compared to other groups. MLT reversed the NP-induced histomorphometric. Also, it changes and increased the activity of superoxide dismutase (SOD), total antioxidant capacity (TAC), and catalase (CAT). The level of malondialdehyde (MDA) significantly decreased in MLT-treated groups compared with the NP group. Conclusion. Our finding showed that MLT enhanced the learning process and reduced NP-induced testicular tissue damage through its antioxidants and cytoprotective effects.

Effect of Aqueous Extract of Chicory (Cichorium intybus L.) Root on Sperm Parameters, Testicular Tissue and Testosterone in Adult Mice Exposed to Diazinon

Results: DZ showed a significant effect on all parameters of sperm, testicular tissue and testosterone (P<0.01). DZ reduced sperm viability by 62% but pretreatment with 100 mg/ kg chicory extract reduced approximately 24% of DZ effects. Straight moving decreased and zig-zag moving, vibrating and non-motile motility increased in sperms. The greatest effect of DZ on sperm morphological abnormalities was increased sperms with tail malformations. Exposure to DZ decreased the sperm count by more than 70%. In addition, weight, size, Leydig, spermatogonia, primary spermatocyte and spermatid cells and serum levels of testosterone were decreased (P<0.01). Chicory extract at the dose of 100 and 200 mg/kg (P<0.01) reduced the effects of DZ but on increasing the dose, its detoxifying effect is reduced.Conclusions: Exposure to DZ reduced male fertility. Chicory extract at a concentration of 100 mg/kg can reduce the detoxifying effect of DZ.
